摘  要:目的探讨前列腺素E2受体(EP1,EP2,EP3和EP4)在人甲状腺乳头状癌(PTC)中的表达情况,并进一步了解EP4下游主要信号分子在人PTC中的表达情况。方法 RT-q PCR法检测15例PTC患者甲状腺癌组织PTC及癌旁正常组织(ANT),和15例结节性甲状腺肿(NG)患者甲状腺组织中的EP1、EP2、EP3和EP4的mRNA表达水平,免疫组化的方法分析EP4及其下游信号分子PKA、Epac和AMPK的蛋白表达水平。结果与ANT及NG组相比,PTC组EP4的mRNA及蛋白水平明显升高(P<0.05),其下游因子PKA、Epac和AMPK的蛋白表达水平也明显升高(P<0.05)。结论 EP4及其下游因子PKA、Epac和AMPK在PTC组织中表达增加,可能参与了PTC的发生及发展。Objective To investigate the expression of prostaglandin E2 receptor(EP1,EP2,EP3 andEP4)in papillary thyroid carcinoma. To understand the expression of EP4 and its downstream signaling pathwayin papillary thyroid carcinoma. Methods RT-q PCR was used to detect the m RNA expression of EP1,EP2,EP3 and EP4 in thyroid tissue of 15 patients with thyroid papillary carcinoma(PTC)and adjacent normal tissues(ANT)and 15 patients with nodular goiter(NG). Then immunohistochemical analysis was used to show theprotein localization and distribution of EP4 and its downstream signaling molecules PKA,Epac and AMPK.Results Compared with ANT and NG group,the m RNA and protein levels of EP4 in PTC group were significantlyincreased(P〈0.05)and the expression levels of PKA,Epac and AMPK were significantly increased(P〈0.05).Conclusion The expression of EP4 and its downstream factors PKA,Epac and AMPK in thyroid tissue ofpatients with thyroid papillary carcinoma were higher and them may be involved in the occurrence and developmentof PTC.
