摘 要:目的比较基于线粒体为靶点的抗氧化剂(MitoQ10)和二甲双胍对多囊卵巢综合征(PCOS)大鼠胰岛素抵抗(IR)的生殖内分泌紊乱的治疗作用,为MitoQ10进一步应用于PCOS-IR治疗的临床研究提供理论依据。方法选取50只21日龄SPF级雌性SD大鼠,随机分为A、B、C、D、E组,使用丙酸睾丸酮和高脂饮食建立PCOS-IR老鼠模型。建模成功后,A组大鼠给予100mg/kg的二甲双胍治疗,B组大鼠给予2mg/mL的MitoQ10治疗,C组大鼠给予100mg/kg二甲双胍和2mg/mL的MitoQ10联合治疗,D组大鼠设定为模型组,E组大鼠设为空白组。干预前后采血测定睾酮(T)、促黄体生成素(LH)、卵泡刺激素(FSH)、空腹胰岛素(FIN)、空腹血糖(FPG),甘油三酯(TG),胆固醇(TC),低密度脂蛋白(LDL),并计算HOMA-IR的指数,最后剖取卵巢进行病理切片观察。结果治疗前后大鼠的血FINS、T、TG、TC、LDL以及HOMA-IR均显著降低(P<0.05);MitoQ10与二甲双胍能在不同程度的改善PCOS-IR大鼠的内分泌代谢的病理改变。结论 MitoQ10可以较好的改善PCOS-IR大鼠的内分泌状态,具有较好的疗效。Objective:To investigate the therapeutic effects of the combination of MitoQ10 and metformin to an animal model of PCOS-IR,and providing the valuable information for treatment of PCOS-IR. Methods:50 SD SPF rats are randomly divided into 5 groups:A,B,C,D and E. Using testosterone propionate and high fat diet,we established a mouse model of PCOSIR. Animals of the group A are administered metformin at concentration of 100 mg/kg,animals of the group B are administered MitoQ10 at concentration of 2 mg/mL,animals of the group C are administered both 100 mg/kg metformin and 2 mg/mL MitoQ10,animals of the group D are designed to be model group,while the group E animals are designed to be control group. Blood are collected to test the testosterone(T),luteinizing hormone(LH),follicle-stimulating hormone(FSH),fast insulin(FIN),fast glucose(FGP),TG,TC,LDL and then calculate the HOMA-IR. Ovarian tissues are collected to evaluate the effect of the treatments. Results:The levels of FINS,FPG,T,TG,TC,LDL and HOMA-IR in these groups animals are decreased significantly after the treatments of metformin and MitoQ10(P〈0.05),suggesting that the MitoQ10 and metformin may be useful for treatment of PCOS-IR. Conclusions:MitoQ10 and metformin may improve the endocrine status of PCOS-IR,which has the therapeutic effects.
