摘  要:目的:建立HPLC-ELSD法分析栀子油中亚麻酸、亚油酸、棕榈酸、油酸和硬脂酸5种高级脂肪酸,并测定其含量。方法:色谱柱Agilent ZORBAX XDB-C18(4.6mm×250mm,5μm),流动相:甲醇-1%乙酸水溶液梯度洗脱,流速:1m L/min,柱温:25℃,ELSD参数:载气:氮气,压力:40Psi,漂移管温度70℃,增益:200,进样量:10μL。结果:亚麻酸、亚油酸、棕榈酸、油酸和硬脂酸的回收率均位于为98.5%~100.5%,相对标准偏差(n=6)分别为1.6%、1.2%、0.8%、1.6%、1.1%。结论:该方法简捷、精确、重复性好,适合栀子油中5种高含量脂肪酸成分的含量测定。Objective: To establish HPLC-ELSD method for analyzing 5 fatty acids in gardenia oil, including linolenic acid, linoleic acid, palmitic acid, oleic acid and stearic acid, and measure the content. Methods: The Agilent ZORBAX XDB-C18 analytical column(4.6 mm×250 mm, 5μm) with methanol-acetic acid aqueous solution as mobile phase at a flow rate of 1.0 mL/min were applied. The column temperature was set at 25℃. The parameters of ELSD were as follows: the carrier gas pressure was 40 Psi, the temperature of the drift tube was 70℃, the plus was 200, sample size was 10μL. Results: The recoveries of linolenic acid, linoleic acid, palmitic acid, oleic acid and stearic acid were all located at 98.5% to 100.5%, and the relative standard deviation(n=6) was 1.6%, 1.2%, 0.8%, 1.6%, 1.1%. Conclusion: This method is simple, accurated and reproducible, it is suitable for the simultaneous determination of the five kinds of high fatty acids in gardenia oil.
