摘 要:目的探讨单羧酸转运蛋白-1(MCT1)与同张力蛋白同源在人第10号染色体上缺失与张力蛋白同源的磷酸酯酶基因(PTEN)在脑胶质瘤中的表达及相关分析。方法采用免疫组织化学二步法免疫组织化学技术(Envision)法及反转录.聚合酶链反应(RT-PCR)法分别检测52例脑胶质瘤织和16例正常脑组织标本中MCT1、PTEN蛋白及mRNA的表达。结果(1)对照组、低级别胶质瘤组、高级别胶质瘤组标本中MCT1、PTENmRNA的表达分别为依次增加和依次下降,差异有统计学意义(P=0.002、0.003);(2)对照组、低级别胶质瘤组、高级别胶质瘤组标本中MCT1、PTEN蛋白表达的阳性率分别为依次增加、依次下降(MCTI:18.8%、61.8%、72.2%;PTEN:68.8%、44.1%、22.2%),差异有统计学意义(P=0.031,0.037);(3)MCT1 PTEN mRNA在脑胶质瘤中的表达无明显相关性(r=0.06,P=0.041)。结论MCT1、PTEN的表达与脑胶质瘤的恶性分级关系密切,病理级别越高MCT1表达呈现显著增强趋势而PTEN则呈现下降趋势。MCT1在胶质瘤中的过表达及PTEN的表达缺失可能都对脑胶质瘤的病理进展起了促进作用,PTEN、MCT1在胶质瘤中的表达无明显相关。Objective To investigate the mRNA and protein expression of monocarboxylate transporters 1 (MCT1) , phosphatase and tensin homologue deleted on chromosome ten (PTEN) in human glio- ma. Methods The mRNA and protein expression of MCT1, phosphatase and tensin homologue deleted on chromosome ten (PTEN) in 52 cases gliomas and 16 normal brain tissus were detected by using reverse transcriptase - polymerase chain reaction ( RT - PCR) and the two - step immunohistoehemieal staining method. Results ( 1 ) The mRNA expressions of MCT1 and PTEN in normal brain tissues, A group ( low grade tissues) , B group (high grade tissues) were increased and decreased in sequence, with significant difference (P = 0. 002, 0. 003) ; (2) In normal brain tissues, A group ( low grade tissues) and B group (high grade tissues) , the poisitive rate of MCT1 protein expression was 18.8% , 61.8% , 72.2% , with significant difference (P =0. 031 ) ; and PTEN in that protein expression was 68.8% , 44. 1% , 22.2% , with signifieant difference ( P = 0. 037 ) ; (3) There have no obvious relationship between MCT1 and PTEN in gliomas (P = 0. 041 ). Conclusion The expression of MCTI and PTEN have a close relationship with the gliomas and it' s malignant degree. The expressions of MCT1 maybe have a positive correlated, But the FTEN maybe as an opposite effects in gliomas. They all may play an important roles together for diagnosis and prognosis in gliomas.
