摘 要:目的:从PI3K/Akt信号传导通路角度探讨安胃汤对慢性萎缩性胃炎模型大鼠疗效的作用机制。方法:采用甲基硝基亚硝基胍(MNNG)慢性萎缩性胃炎大鼠模型,应用安胃汤进行干预;运用HE染色观察安胃黏膜组织形态改变;采用实时荧光定量PCR方法、Western blot检测PTEN、PI3K、Akt、PDK1、XIAP基因表达及其蛋白含量的水平。结果:安胃汤能够明显改善CAG大鼠胃组织的病理变化,该方可以明显上调PTEN基因以及其蛋白含量的表达,下调PI3K、Akt、PDK1、XIAP基因以及其蛋白含量的表达。结论:安胃汤能够明显改善CAG大鼠胃组织的病理变化,具有明显的抗CAG作用,其作用机制可能通过提高PTEN基因及蛋白的表达,降低PI3K、PDK1、Akt基因及蛋白表达,抑制CAG大鼠胃黏膜细胞P13K/Akt信号传导通路,降低XIAP基因及蛋白,促进胃黏膜细胞凋亡,可能是安胃汤防治CAG的关键机制之一。Objective: To explore the mechanism underlying the effects of Anwei Decoction on model rats with chronic atrophic gastritis(CAG) from the perspective of the PI3 K/Akt signalling pathway. Methods: MNNG was used to establish the rat model of CAG which was then subject to the intervention by Anwei Decoction. HE staining was used for observation of morphological change in gastric mucosa. The quantitative real-time fluorescent PCR and Western blot were used to measure the expressions of genes PTEN,PI3 K,Akt,PTEN,PDK1 and XIAP and their proteins. Results: Anwei Decoction is able to obviously improve pathological changes of gastric mucosa of CAG rats. The formula significantly up-regulated the expressions of PTEN and its proteins,and down-regulated the expressions of genes PI3 K,PDK1,Akt and XIAP and their proteins. Conclusion: Anwei Decoction is able to obviously improve pathological changes of gastric mucosa of CAG rats and has an obvious anti-CAG effect. Such effects may be produced by increasing the expression of PTEN and its proteins,decreasing the expressions of PI3 K,PDK1 and Akt and their proteins,inhibiting the PI3 K/Akt signaling pathway,reducing the expression of XIAP and its proteins,and promoting apoptosis of gastric mucosal cells in gastric mucosal cells of CAG rats.
