摘  要:为了评估表达产气荚膜梭菌NetB毒素的重组植物乳酸杆菌口服活菌疫苗对鸡的免疫保护效果,分别进行了重组活菌疫苗的免疫和产气荚膜梭菌攻菌试验。设置3组免疫组雏鸡,于7~28日龄分别连续免疫pSIP409空载体植物乳酸杆菌、pSIP409-Net B重组植物乳酸杆菌和灭菌PBS;设置4组攻菌组雏鸡,免疫方法同上,免疫后于43~47日龄连续口服产气荚膜梭菌(设置PBS免疫组不攻菌只口服等量PBS对照)。通过免疫后肠道分离重组菌、病理组织学检查疫苗对雏鸡组织器官有无损害作用及保护作用;攻菌后细菌分离计数、剖检肠道病变评分来评价疫苗的免疫水平。结果显示,免疫后2周仍可在免疫鸡的小肠、盲肠段分离到重组菌;组织学检查发现与对照组相比重组疫苗的免疫没有对雏鸡组织器官产生损害作用;NetB攻菌组的肠道损伤情况较空载体攻菌组和PBS攻菌组轻微;NetB攻菌组的肠道病变评分低于空载体攻菌组和PBS攻菌组。表明所构建的重组口服活菌疫苗安全有效,结果为重组植物乳杆菌表达系统作为口服疫苗应用提供了理论依据。To evaluate the protective effect of recombinant Lactobacillus plantarum oral vaccine with NetB toxin of Clostridium perfringens in chickens, the recombinant live vaccine immunization and Clostridium perfringens challenge test were carried out respectively. The immune treatment was divided into 3 groups at 7 to 28 days of age continuously inoculate pSIP409 empty vector Lactobacillus, pSIP409-NetB recombinant Lactobacillus and sterile PBS, respectively.Immune-challenge group treatment was divided into 4 groups, the immunization method was the same as above, which was continuously oral inoculated C. perfringens at 43 to 47 days of age. Recombinant bacteria isolated from the intestine and histopathological examination were used to inspect the protective or damage effect of the vaccine for chicken tissues and organs. bacteria isolate and count and intestinal cesarean section and lesion score were used to evaluate the overall immunity of the vaccine. The results showed that the recombinant bacteria could be isolated from the small intestine and the cecum segment of the chicken in 2 weeks. Histological examination showed that compared with the control group, the immunization of recombinant vaccine did not damage the tissues and organs of chickens. The intestinal injury of NetB group was lighter than that of empty vector group and PBS group. Intestinal lesion score of Net B group was lower than that of empty vector group and PBS group. Ihe results indicated that the recombinant vaccine was safe and effective,and could provide a theoretical basis for the application of recombinant expression system in Lactobacillus oral vaccine.
