摘 要:目的分析尿液标本不同保存方法对24h尿蛋白检测结果的影响,探讨留取24h尿液适宜的保存方法。方法在同一时间收集尿液后,分别加入防腐剂和不添加防腐剂,在不同保存温度、不同保存时间的条件下,采用单因素方差分析防腐剂、温度、保存时间三种因素对24h尿蛋白检测结果的影响。结果不论尿液中是否加入二甲苯防腐剂,在2℃-8℃、18℃~28℃以及37℃三种温度条件下保存24h后,各组24h尿蛋白检测结果差异均未见统计学意义(P〉0.05);加有二甲苯防腐剂的尿液标本,在2℃~8℃、18℃~28℃以及37℃三种温度条件下分别保存24、48、72h后,各组24h尿蛋白检测结果差异均未见统计学意义(P〉0.05);无防腐剂的尿液标本,在2℃~8℃务件下分别保存24、48、72h后,24h尿蛋白检测结果差异均未见统计学意义(P〉0.05),在18℃~28℃以及37℃条件下分别保存24、48、72h后,24h尿蛋白检测结果稳定性分别保持在48h和24h,差异有统计学意义(P〈0.05),且2℃-8℃条件下,尿蛋白检测结果的稳定性显著优于18℃-28℃以及37℃的保存条件。结论24h尿蛋白测定的标本可以在无二甲苯防腐剂条件下进行室温保存,该方法安全、简便、可行,建议在临床上推广应用。Objective To analyze the effect of different preservation methods of urine on the result of 24 hours urine protein, and in order to find suitable preservation method for 24 hours urine. Methods Urine samples were collected at the same time and added antiseptic or without respectively. The effects of antiseptic, temperature and storage time on the detection of 24 hours urine protein were analyzed by one-way ANOVA at different storage temperature and time. Results Whether urine samples were added xylene or without, there was no significant difference in the 24 hours urine protein at the temperature of 2℃ -8℃, 18℃ -28℃ and 37℃ after storage for 24 hours (P 〉0.05). At the temperature of 2℃ - 8℃, 18℃ - 28℃ and 37℃, there was no significant difference in the results of 24 hours urinary protein among all groups of added xylene after storage at 24 hours, 48 hours and 72 hours respectively (P 〉 0. 05). At the temperature of 2 ℃ - 8℃, there was no significant difference in the results of 24 hours urinary protein among the groups without xylene after storage in 24 hours, 48 hours and 72 hours (P 〉0. 05). At the temperature of 18 -28℃ and 37℃ and storage in 24 hours, 48 hours and 72 hours, the stability of the 24 hours urine protein result was kept at 48 hours and 24 hours respectively, there was significant difference (P 〈 0. 05 ). Under the condition of 2℃- 8℃, the stability of the 24 hours urine protein detection results were significantly better than those at 18℃ -28 ℃ and 37℃. Conclusions Samples of 24 hours urinary protein can be stored at room temperature without xylene preservative. The method is safe, simple and feasible, which may be recommended to application in clinics.
