摘 要:目的:探讨脂多糖对食管鳞状上皮细胞的骨形态蛋白4(BMP4)表达的影响。方法:体外培养正常食管鳞状上皮Het-1A细胞,采用不同浓度脂多糖(Lipopolysaccharide,LPS)处理细胞、实时荧光定量PCR与蛋白印迹分析方法测定BMP4及Smad1表达的变化,并对其进行相关性分析。结果:10 ng/mL LPS组可轻微刺激食管鳞状上皮细胞表达BMP4、Smad1的表达,与对照组比较差异均无统计学意义(P>0.05);100 ng/mL LPS组和500 ng/mL LPS组均明显增强BMP4和Smad1的表达,且随浓度增强效应增强,差异均有统计学意义(P<0.05)。结论:脂多糖可能通过诱导BMP4及BMP信号通路相关蛋白Smad1的表达参与Barrett食管的发生。Objective:To explore the effect of Lipopolysaccharide on BMP4 expression in esophagus squamous cells.Method:Primary Het-1 A cells were treated with Lipopolysaccharide at different concentration.The changes of expressions of BMP4 and Smad1 were examined by real time polymerase chain reaction and Western blot analysis.Result:10 ng/mL LPS group could slightly stimulate the expression of BMP4 and Smad1 in the squamous epithelial cells of the esophagus,there was no significant difference compared with the control group(P〈0.05).The expressions of BMP4 and Smad1 were obviously enhanced in the 100 ng/mL LPS group and the 500 ng/mL LPS group,and the difference was statistically significant with the enhancement of the concentration enhancement effect(P〈0.05).Conclusion:Lipopolysaccharide can regulate the expressions of BMP4 and Smad1,which may play a role in the development of Barrett oesophagus.
