摘 要:目的建立HPLC-UV测定红参中人参皂苷Rg_1、Re、Rf、Rb_1、Rc、Rb_2含量测定方法,以及Rb_3和Rd的定性鉴别方法,用于区别国产红参与进口红参。方法采用YMC-Pack ODS-A(4.6 mm×100 mm,3μm)色谱柱,以乙腈-0.1%磷酸为流动相梯度洗脱,流速0.6 mL·min^(-1),检测波长203 nm,柱温30℃。结果方法学验证结果良好,并采用t检验和主成分分析方法对测定数据进行分析,得到区分高丽红参和国产红参的主要因素为人参皂苷Rg_1和Rd。结论确定人参皂苷Rg_1和Rd峰面积比为区分二者的质控指标,为区分高丽红参与国产红参提供了可行的方法。OBJECTIVE To establish an HPLC-UV method for the quantitative determination of ginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2 and the qualitative determination of ginsenosides Rb3 and Rd in Red Ginseng. This method is used to make different between the imported Red Ginseng and China Red Ginseng.METHODS The analysis were performed on a YMC-Pack ODS-A column(4.6 mm×100 mm,3 μm), the mobile phase was acetonitrile -0.1% phosphoric acid at the flow rate of 0.6 mL·min-1, the detection wavelength was set at 203 nm,the column temperature was maintained at 30 ℃.RESULTS The method herein is effective. The data of samples was subjected to t test and principal component analysis(PCA)in order to find the marker constituents. According to the information of t test and PCA,ginsenoside Rg1 and ginsenoside Rd were the main factor to classify Korean Red Ginseng and Chinese Red Ginseng.CONCLUSION The peak area ratio of ginsenoside Rg1 to ginsenoside Rd is used as the quality control parameters. This method is suitable to classify Korean Red Ginseng and Chinese Red Ginseng.
