摘  要:目的通过海藻多糖(SP)与锌离子(II)络合成一种新型的海藻多糖锌(SPZC),在比较SP和SPZC二者理化性质、结构组成基础上,对锌离子的多糖螯合物在单核细胞THP-1细胞耗氧的影响做了初步的研究。方法:用苯酚法法测定SP和SPZC的总多糖含量、Hodgson法测定二者的硫酸根含量和Folin酚试剂法等化学法测定二者主要作用成分,气相色谱(GC)分析了二者所含的主要单糖及摩尔比,红外光谱(IR)分析多糖二者的主要成分基团等。用ESR自旋捕捉法检测了二者对细胞的呼吸爆发作用(respiratory burst,RB)下耗氧量的影响,以及利用活性氧(reactive oxygen species,ROS)的荧光探针(2,7-二氢二氯荧光素,DCFH-DA)分析了PMA激活THP-1细胞后ROS产生的变化。结果 SP络合上锌离子成为SPZC后也具有很好的稳定性,二者之间的主要成分含量变化明显,多糖含量、蛋白质含量、硫酸根离子下降明显;而SPZC中Zn(II)的含量为15%。通过GC分析,表明锌离子的配位反应,使得SP的单糖组成也有改变;IR图谱可以发现SP结合上锌离子后,图谱上的特征性峰发生位移、钝化或消失,而代之有SPZC核的类型峰。RB结果,提示在细胞受到激发的短暂时间内,SPZC比SP更能维持细胞的RB在一个更高水平;而同样细胞激发后的较长时间内,ROS产生的荧光染色,相较SP对ROS产生有一定的淬灭作用,而SPZC则能触发ROS产生更多。结论 SP同锌离子(II)络合产生的SPZC,增加了新的有机锌来源;同时,SPZC对比SP,其抗氧化性能有所减弱。Objective To synthesize seaweed polysaccharides zinc complex(SPZC) from Zn2+ and seaweed polysaccharides(SP) which were extracted from seaweed, and compare its main component, and activity on oxygen consumption, generation of reactive oxygen species(ROS) in THP-1 cells with SP. Methods The functional component was measured with physiol-chemical methods, main groups by Infrared Spectroscopy(IR), and main monosaccharide by gas chromatography(GC), respiratory burst(RB) by self-spinning ESR technique and ROS by 2',7'-dichloro fluorescein(DCFH-CA) staining, respectively. Results The outcome indicated that both SP and SPZC have good stability, but main contents of total polysaccharides, sulfate ions, protein, and Zn2+ concentration were different. The investigation by infrared spectra indicated that the main group of SPZC had been shifted, inactivated or disappeared based on the IR of SP. Both could lift the RB at a high level while SPZC had been more effective than SP in THP-1 cells after PMA stimulation within a short period. As to the ROS generation, SPZC was highly active in maintaining stronger ROS fluorescence extensity,while SP was a little bit effective in weakening the fluorescence extensity. Conclusion The SPZC synthesized from SP and Zn2+ is a new source of organic zinc products. However SPZC seems stronger in activating RB and ROS generation.
