摘 要:为提高马铃薯抗逆性,利用农杆菌介导法,将携带目的基因HaNAC1质粒分别导入马铃薯栽培种‘大西洋’、‘费乌瑞它’和‘青薯168’中。对卡那霉素(Kan)抗性转化株系经PCR扩增检测,共得到63个阳性株系。利用Southern blot对随机选取的7个转基因阳性株系进行检测,结果表明HaNAC1均已整合到各转基因株系中。对63个阳性株系进行RT-PCR检测,共有55个株系中出现目的基因条带,说明HaNAC1在转录水平有表达。以90 mmol/L NaCl和15%PEG分别添加至培养基中模拟盐和旱胁迫处理,从相对株高、相对根长等形态指标对32个‘大西洋’转基因株系进行筛选获得了4个优系,丙二醛、脯氨酸等生理指标也进一步证实HaNAC1基因转化可有效提高转基因马铃薯的抗逆性。In order to enhanced the stress tolerance of potato,the plasmid encoding HaNAC1 gene was transferred into cultivars of Atlantic,Favorita and Qingshu 168 respectively via Agrobacterium tumefaciens mediated method.After PCR amplification detection,63 positive transgenic lines were obtained from Kanamycin resistant lines.7 positive lines were chosen randomly for Southern blot analysis,the results of which confirmed that HaNAC1 gene had been integrated into the potato.The RT-PCR analysis of 63 positive lines showed that 55 lines had positive target bands,indicating that HaNAC1 expressed at the transcription level.90 mmol/L NaCl and 15% PEG were added to the culture medium to simulate salt and drought stress,and 4 best lines were screened from 32 Atlantic transgenic lines according to relative plant height and relative root length.Malondialdehyde,proline and other physiological indicators further confirmed that HaNAC1 gene transformation could effectively improve the resistance of transgenic potato.
