摘 要:目的探讨神经生长因子(nerve growth factor,NGF)及其受体p75NTR在喉黏膜上皮、不典型增生、乳头状瘤、鳞状细胞癌等组织中的表达、分布情况,并分析其潜在的生物学功能。方法应用免疫组化染色法,针对临床组织病理标本,检测NGF及受体p75NTR在42例喉鳞状细胞癌组织、14例不典型增生黏膜、6例喉乳头状瘤及11例正常喉黏膜组织中的表达。结果喉鳞状细胞癌组织中存在NGF及p75NTR阳性表达。伴有颈部淋巴结转移的喉鳞状细胞癌中NGF表达的强阳性率高于无淋巴结转移组(P<0.05)。p75NTR阳性细胞在喉鳞状细胞癌中呈区域性分布。在中、高分化喉鳞状细胞癌组织中,p75NTR主要表达于生长活跃的癌巢基底层和浸润缘,该区域细胞表现较强的增殖能力;在低分化喉鳞状细胞癌组织中,p75NTR阳性细胞分布范围逐渐分散。结论 NGF表达升高与喉鳞状细胞癌进展及淋巴结转移相关。p75NTR高表达于喉鳞状细胞癌组织中的未成熟细胞和具有较强增殖能力的细胞。NGF及p75NTR的表达对支持和促进喉鳞状细胞癌细胞生长具有重要作用,相关调节机制与喉鳞状细胞癌的发生及进展密切相关。Objective To investigate expressions and distributions of nerve growth factor (NGF) and p75NTR receptor in laryngeal mucous epithelium, atypical hyperplasia, papilloma and laryngeal squamous cell carcinoma ( LSCC ) tissues and to analyze its potential biological function. Methods Expressions of NGF and p75NTR receptor were detected by immunohistoehemieal technique method in 42 tissues of 1SCC, 14 tissues of atypical hyperplasia, 6 tissues of laryngeal mucous epithelium and 11 eases of normal tuniea mueosa ISCC tissues. Results Positive expressions of NGF and p75NTR receptor were found in laryngoeareinoma tissues. Strong positive expression ratio of NGF in laryngoeareinoma patients associated with cervical lymph node metastasis was higher than that in laryngoeareinoma patients without lymph node metastasis (P 〈 0.05 ). Positive p75NTR receptor cell was regionally expressed in ISCC tissues. In moderately and highly differentiated LSCC tissues, p75NTR receptor was mainly expressed in the basilar part and infihrative margin of active cancer nest, at which the cells had high proliferation ability. In poorly differentiated LSCC tissues, distribution of p75NTR receptor positive cells was diffusely gradually. Conclusion Increased NGF expression is related to laryngeal squamous cell carcinoma development and lymph node metastasis, and p75NTR receptor is highly expressed in immature cells and high proliferation ability cells in laryngocarcinoma tissues. NGF and p75NTR expressions have important role in supporting and promoting growth of ISCC cells, and related regulatory mechanism is closely related to carcinogenesis and development of ISCC.
