整合素β1在鼻咽癌的表达与5年生存情况的关系

摘　　要：目的检测整合素β1(integrinβ1,ITGB1)在鼻咽癌组织中的表达,探讨整合素β1的表达意义及与临床预后的关系。方法采用免疫组织化学技术检测60例鼻咽癌组织及10例正常癌旁黏膜组织整合素β1的表达情况(整合素β1染色面积积分+染色强度积分<3分为低表达,≥3分为高表达),分析其与鼻咽癌临床病理特征及预后的关系。结果鼻咽癌组织整合素β1表达阳性率为61.7%,正常黏膜组织仅呈弱阳性表达于基底细胞(0%),两者比较差异有统计学意义(P=0.000)。转移组整合素β1的表达较非转移组明显增高(P=0.031),且整合素β1表达水平与鼻咽癌的T分期、临床分期呈正相关(r=0.301,P=0.024;r=0.267,P=0.039)。整合素β1与性别、年龄、发病部位及淋巴结转移无明显相关性(P>0.05)。整合素β1高表达组鼻咽癌患者5年总生存率明显低于低表达组(P=0.041)。结论整合素β1在鼻咽癌组织的表达明显高于正常黏膜组织,整合素β1表达水平与肿瘤远处转移、T分期及临床分期密切相关。Objective To investigate the expression of integrinβ1（ ITGB1） in nasopharyngeal carcinomas（ NPC）,and to investigate the significance of ITGB1 and its correlation with prognosis. Methods Using immunohistochemical staining,the expression of ITGB1 was detected in 60 cases of NPC and 10 cases of normal mucosa adjacent to the carcinoma. The correlation between ITGB1 and clinicopathological features and prognosis were analyzed. Results The positive rate of ITGB1 in NPC tissues were 61. 7%,yet it was weakly positive in basal lamina cells of normal mucosa. And there was a significant difference between the two groups（ P = 0. 000）. Comparing with non-metastatic patients,the level of ITGB1 was significant higher in metastatic patients（ P = 0. 031）. ITGB1 was significantly positively correlated with T grades and clinical stage of NPC（ r = 0. 301,P = 0. 024; r = 0. 267,P = 0. 039）. No significant difference was observed between ITGB1 and sex,age,location,or lymph node metastasis（ P 〉 0. 05）. The total 5-year survival rate of the NPC patients with high expression of ITGB1 was significantly lower than that of the low expression group（ P = 0. 041）. Conclusion The level of ITGB1 in NPC tissues is significantly higher than that in normal mucosa,and is closely correlated to distant metastasis,T stage and clinical stage.
