摘 要:[目的]分析四倍体马铃薯"合作88"PVY、PLRV抗性基因的基因型。[方法]利用969个自交分离群体株系,采用田间性状调查、DAS-ELISA病毒检测、抗性基因连锁分子标记检测。[结果]田间调查PVY、PLRV抗病与感病的比为968.000∶1和41.130∶1,推测基因型分别为YYYy和RRrr,分离方式为染色单体随机分离和染色体随机分离;DAS-ELISA检测PVY、PLRV阴性与阳性的比为483.500∶1和19.617∶1,推测基因型分别为YYYy和RRrr,分离方式均为完全均衡式分离;分子标记RYSC3检测PVY抗性基因Ryadg有特异性和无特异性的比为322.000∶1,推测PVY基因型为YYYy,分离方式为完全均衡式分离;分子标记DMB32-11检测PLRV抗性基因Rlretb有特异性和无特异性的比为32.414∶1,推测PLRV基因型为RRrr,分离方式为染色体随机分离。[结论]3种试验方法均表明四倍体马铃薯"合作88"PVY抗性基因的基因型是YYYy,PLRV抗性基因的基因型是RRrr,二者在遗传分离时存在多种分离方式。[Objective] To analyze the genotype of PVY and PLRV resistance genes in tetraploid potato‘Cooperation-88'. [Method] In this study,field investigation,DAS-ELISA assays and resistance gene linked molecular marker detection were used to analyze 969 self-inbred lines. [Result] In field investigation,the ratios of PVY and PLRV resistance and susceptibility plants were 968. 000 ∶ 1 and 41. 130 ∶ 1,respectively. It was speculated that the resistance genotypes of PVY and PLRV were YYYy and RRrr,and the corresponding separation ways were chromatids random separation and chromosomes random separation. The DAS-ELISA detection results showed that the PVY and PLRV infection ratios between negative and positive were 483. 500∶ 1 and 19. 617∶ 1. The deduced resistance genotypes of PVY and PLRV were YYYy and RRrr and they all run the complete equilibrium segregation way. The PVY resistance gene,Ryadg,had a genotype of YYYy and a ratio of322. 000 existence to 1 deficiency in self inbred lines according to the detection of PVY linked molecular marker RYSC3,segregated in a complete equilibrium segregation way. Meanwhile,PLRY linked molecular marker DMB32-11 was used to check the ratio between the existence or deficiency of PLRV resistance gene Rlretb. The ratio was 32. 414∶ 1. The result showed Rlretbmay has a genotype of RRrr with the way of random chromosome segregation. [Conclusion] Three methods were used to investigate genotypes of PVY and PLRV resistance genes in potato cultivar ‘Cooperation 88'. Each method can deduce to the same genotypes of PVY and PLRV( YYYy and RRrr),and two potato virus genes shared several segregation ways during genetic reproduction.
