摘　　要：本研究旨在探讨甲状腺腺瘤患者Wnt/β-catenin信号通路相关蛋白Wnt-1,β-catenin的检测及临床意义。采用RT-PCR方法检测Wnt-1,β-catenin mRNA表达情况,采用Western blot和免疫组化方法检测Wnt-1,β-catenin蛋白表达情况。结果显示,与对照组相比,疾病组Wnt-1,β-catenin mRNA和蛋白的表达量均升高。肿瘤组织中,疾病组的Wnt-1蛋白表达阳性率和β-catenin蛋白表达阳性率均高于对照组。结果表明,Wnt-1表达与甲状腺腺瘤患者肿瘤直径密切相关。β-catenin表达也与甲状腺腺瘤患者肿瘤直径密切密切相关。甲状腺腺瘤患者Wnt/β-catenin信号通路相关蛋白Wnt-1,β-catenin蛋白表达上调,且其表达与肿瘤生物学状态密切相关。The aim of this study was to evaluate the expressions of Wnt/β-catenin signal pathway related protein in thyroid cancer and their clinical significance. Quantitative real-time PCR was performed to assess mRNA levels of Wnt-1,β-catenin. Western blot assays and immunohistochemistry were performed to assess protein levels of Wnt-1,β-catenin. The results showed that,compared with the control group,the expression of Wnt-1,beta-catenin mRNA and protein increased in the disease group. In the tumor tissue,the positive rate of Wnt-1 protein expression and the positive rate of the expression of beta-catenin protein in the disease group were higher than those in the control group. The results showed that the expression of Wnt-1 was closely related to tumor diameter in patients with thyroid adenoma. The expression of beta-catenin is also closely related to tumor diameter in patients with thyroid adenoma. The expressions of Wnt/β-catenin signaling pathway related proteins Wnt-1,β-catenin are increased in patients with thyroid cancer,which may play an important role in the occurrence and development of thyroid cancer.
