摘  要:目的:探讨性激素对子宫肌瘤的发生和生长的调节作用。方法:采用体外子宫内膜间质细胞培养,加以雌二醇(E2)、孕酮(P)、睾酮(T)刺激后,进行流式细胞学测定其细胞周期和凋亡的动力学变化。结果:T单独作用时其促进DNA合成及有丝分裂效果最明显,但与E2或P合用时其作用大大削弱;而E2+P对细胞DNA合成及有丝分裂的促进作用比单用E2强,两者的这种协同作用在有T存在的情况下也被减弱。P对细胞的G2+M期作用最强。T对细胞凋亡具有促进作用,T和P之间存在拮抗作用,P能够抑制细胞凋亡。结论:雌孕激素与雄激素之间的代谢平衡对于维持细胞的正常功能至关重要,若局部组织细胞增生与凋亡失衡则会导致肿瘤的发生。Objective:To investigate the actions of ovarian steroids in the regulation of proliferation and apoptosis of human uterine leiomyoma.Methods:Stromal cells derived from proliferative human endometrium have been isolated,the populations of endometrial stromal cells exhibited over95%homogeneity,ascertained immunocytochemically,and were cultured in the presence of estradiol(E2),testosterone(T)and progesterone(P),then flow cytometry was examined.Results The cells exposed to T alone were induced DNA synthesis and mitogenesis most obviously,but T in combination with E2or P decreased this effect greatly.E2+P induced the cells DNA synthesis and mitogenesis more effective than E2alone.However,this synergic effect was dampened in the presence of testosterone.Progesterone stimulated the cells into G2+M phase most effectively.Testosterone promotes apoptosis of human ESC and P suppresses apoptosis.Conclusion These results suggest that the proportional balance of steioroids hormones in the local tissues is important to keep normal physiological functions of the cells.Thus,we speculated the imbalance of proliferation and apoptosis of the cells in the local tissues may be the mechanism of tumor genesis.
