摘  要:目的观察不同浓度菝葜皂苷元对体外培养的胃癌BGC-823细胞凋亡相关蛋白Bax、Bcl-2表达的影响。方法采用免疫组化方法,检测BGC-823细胞经不同浓度(12.5、25、50μg/mL)的菝葜皂苷元处理后Bcl-2、Bax的表达情况。结果随着菝葜皂苷元浓度的增加,与对照组比较,BGC-823细胞Bax表达逐渐增加,不同浓度组之间差异有统计学差异(P<0.05),而Bcl-2表达逐渐减少(P<0.05)。结论菝葜皂苷元可诱导BGC-823细胞凋亡,上调Bax蛋白水平与下调Bcl-2蛋白水平。Objective To investigate the effects of different concentrations of sarsasapogenin on apoptosis of Bax and Bcl-2 apoptosis-related protein expression of human gastric cancer cell line BGC-823 cultured in vitro. Methods The BGC-823 cells were treated by sarsasapogenin in different concentrations, Flow cytometry was used to assay the effects of sarsasapogenin on apoptosis and immunocytochemical method was used to assay Bax, Bcl-2 protein expression. Results The apoptosis rate of BGC-823 cell were significantly augmented and bax expressions increased and Bcl-2 expressions decreased while the concentration and duration of sarsasapogenin increased(P 0.05) respectively. Conclusion Sarsasapogenin can efficiently induce apoptosis of BGC-823 cell, up-regulate Bax and down-regulate Bcl-2 protein expression which are the possible mechanisms for its anti-cancer effect.
