紫杉醇联合顺铂调控MAPK/NF-κB信号治疗幽门螺杆菌胃癌机制的研究

摘　　要：目的:探讨紫杉醇联合顺铂对幽门螺杆菌胃癌的治疗作用,及丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)/核转录因子kappa B(nuclear factor kappa-light-chain-enhancer of activated B cells,NF-κB)信号通路在幽门螺杆菌胃癌患者中的表达变化及意义。方法:给予幽门螺杆菌胃癌患者紫杉醇联合顺铂化疗2个疗程,采用免疫组织化学法检测胃癌患者癌组织中MAPK和NF-κB蛋白的表达;采用Real-Time PCR法检测胃癌组织中MAPK和NF-κB mRNA的表达;采用Western blotting检测胃癌组织中凋亡蛋白MAPK和NF-κB表达;ELISA法检测患者p65和Caspase 3含量。结果:与治疗前相比,治疗后胃癌患者癌组织中的MAPK和NF-κB蛋白表达及mRNA表达水平显著升高,凋亡蛋白p65和Caspase 3表达也显著性增加,差异均有统计学意义(P<0.05)。结论:紫杉醇联合顺铂对幽门螺杆菌胃癌有一定的治疗作用,且MAPK/NF-κB信号通路在此过程中发挥重要的作用。Objective： To investigate the therapeutic effect of paclitaxel combined with cisplatin on Helicobacter pylori gastric cancer,and the significance of MAPK/NF-κB signaling pathway in patients with Helicobacter pylori（ Hp）-related gastric cancer. Methods： Helicobacter pylori gastric cancer patients given paclitaxel combined with cisplatin chemotherapy 2 course of treatment,and immunohistochemical method was used to detect the expression of MAPK and NF-κB protein in cancer tissues of patients with gastric cancer. The expressions of MAPK and NF-κB mRNA in gastric carcinoma were detected by real-time PCR. Western blotting was used to detect the expression of apoptotic protein MAPK and NF-κB in gastric carcinoma. The levels of p65 and Caspase 3 were detected by ELISA. Results： Compared with before treatment,the expression of MAPK and NF-κB protein and mRNA in cancer tissues of patients with gastric cancer were significantly increased after treatment. The expression of apoptotic protein p65 and Caspase 3 also increased significantly,the difference was statistically significant（ P〈0. 05）. Conclusion： Paclitaxel combined with cisplatin has a certain therapeutic effect on Helicobacter pylori gastric cancer. And MAPK/NF-κB signaling pathway plays an important role in this process.
