摘 要:旨在探索circRNAs在脂肪组织可能发挥的潜在作用,为了解circRNA调控脂肪沉积和脂代谢作用的机制奠定基础。本研究采用RNA-Seq和生物信息学方法鉴定大白和莱芜猪肌内脂肪组织中circRNAs的表达情况,运用miRanda和Targetscan对差异表达circRNA进行靶基因预测,对靶基因进行GO和KEGG富集分析。结果发现,181个差异表达circRNAs得到鉴定,其中,102个在莱芜猪肌内脂肪组织中上调表达,79个下调表达。并对具有较多miRNA结合位点的ssc_circ_0002807、ssc_circ_0009352靶基因进行了GO和KEGG富集分析,发现靶基因富集于胆固醇平衡和磷脂平衡等与脂代谢相关的生物学过程。通过qRT-PCR,验证了ssc_circ_0002807、ssc_circ_0005382、ssc_circ_0009172和ssc_circ_0004824,与测序结果一致。由此推断,ssc_circ_0002807与ssc_circ_0009352可能参与调控脂肪沉积和脂质代谢。The purpose of this study was to explore circRNAs potential role in adipose tissue,which would lay a basis for further understanding the regulation role of circRNA in fat deposition and lipid metabolism.In this study,we explored the expression profile of circRNAs in intramuscular adipose tissue from Large White and Laiwu pigs using RNA-seq and bioinformatics.The target genes to the differentially expressed circRNAs were predicted by miRanda and Targetscan.The enrichment analysis of target genes was performed by GO and KEGG.The results showed that 181 differentially expressed circRNAs were identified.Among them,102 were up-regulated expression in intramuscular adipose tissue of Laiwu pigs,and 79 were down-regulated.GO and KEGG enrichment analysis of target genes of ssc_circ_0002807 and ssc_circ_0009352 with more miRNA binding sites were peformed.The target genes were found to be enriched in the biological process involved in lipid metabolism such as phospholipid homeostasis,cholesterol homeostasis,etc.And ssc_circ_0002807,ssc_circ_0005382,ssc_circ_0009172 and ssc_circ_0004824 were also confirmed by qRT-PCR.It is infered that ssc_circ_0002807 and ssc_circ_0009352 may regulate fatdeposition and metabolism.
