摘  要:目的观察N-对甲苯磺基-L-苯乙胺酰氯甲基酮(N-tosyl-L-phenylalanyl-chloromethyl Ketone,TPCK)干预对延缓晶状体混浊程度的作用,并初步分析TPCK在晶状体氧化应激状态下的保护作用。方法取同窝SD大鼠离体培养晶状体36个,随机分为3组:空白对照组、过氧化氢组(H_2O_2组)和药物干预组(TPCK组),每组12个。于晶状体培养12h显微镜下观察离体培养晶状体混浊的情况并分别计分;然后每组取一个晶状体匀浆进行热稳定性分析;其余所有晶状体匀浆后测定谷胱甘肽(glutathione,GSH)含量。结果TPCK组晶状体混浊程度(8.33%)明显低于H_2O_2组(83.33%),差异有统计学意义(Z=-3.862,P<0.05)。TPCK组GSH含量为(2.36±0.29)μmol·g^(-1),与H_2O_2组(1.12±0.08)μmol·g^(-1)相比明显升高,低于空白对照组(3.52±0.30)μmol·g^(-1),差异有统计学意义(F=232.89,P<0.05)。结论 TPCK作为一种烷基化试剂竞争性结合晶状体蛋白巯基而减轻了其氧化损伤程度。TPCK能稳定晶状体蛋白的热变性。Objective To observe the role in N-tosyl-L-phenylalanyl-chloromethyl Ketone (TPCK) on postponing crystal turbidity and analyze the effect of TPCK in crystalline oxidative damage.Methods Together 36 intact lenses were cultured and randomly divided into 3 groups:normal control group,H2O2 group,and TPCK group (treatment group),and each group had 12 objects.The crystal turbidity and the opacification level were observed after 12 hours.Crystalline protein thermal stability changes of one lens taking out of each group were analyzed,and then the level of glutathione (GSH) in the remaining lens of each group were measured.Results The lens opacification level in the TPCK group (8.33%) was obviously lower than that in the H2O2 group (83.33%).The difference was statistical significance (Z=-3.862,P〈0.05).The GSH level in the TPCK group (2.36±0.29)μmol·g-1 was significantly higher than that in the H2O2 group (1.12±0.08)μmol·g-1,but lower than that in the control group (3.52±0.30)μmol·g-1(F=232.89,P〈0.05).Conclusion TPCK as an alkylating agent competitively binds to the lens protein sulfhydryl group to reduce the degree of oxidative damage.TPCK stabilizes the thermal denaturation of lens proteins.
