摘 要:建立三七面霜中3种皂苷(三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1)总含量的近红外光谱快速测定方法。采用乳化工艺制备含三七提取物的面霜,采用HPLC法测定30份三七面霜中3种皂苷的总含量。采集三七面霜在12 000~4 000 cm-1的NIR光谱,分别对建模波段和光谱预处理方法进行优选,确定最佳主成分数,采用消除常数偏移量法建立三七面霜中3种皂苷总含量与近红外光谱之间的校正模型。HPLC法测定三七面霜中3种皂苷的总含量为0.6~4.11 mg/g;三七面霜的近红外光谱模型预测波长为6 102.1~5 446.4 cm-1,主因子维数选择4,预测集的相关系数R2为0.9375、校正均方根偏差RMSEC为0.451、预测均方根偏差RMSEP为0.588、相对分析误差RPD为4.69;平均预测回收率为103.33%±11.07%。所建立的模型性能较好,可用于三七面霜中3种皂苷总含量的快速检测。To establish the rapid quantitative determination method of the total content of three kinds of saponins( notoginsenoside R1,ginsenoside Rg1 and ginsenoside Rb1) in Panax notoginseng cream by near-infrared spectrum( NIR),total saponins were extracted from Panax notoginseng with water,the 30 batches of Panax notoginseng cream were prepared by emulsification technique.The contents of three kinds of saponins in cream were determined by HPLC. The samples of Panax notoginseng cream in the NIR spectrum of 12 000 ~ 4 000 cm^-1 were collected. The optimal principal component number was determined by optimizing the model parameters including pretreatment method of spectra and wavelength range spectively. The eliminate constant offset method was used to establish the calibration models between the spectra and the total contents of main saponins. The total content of the three kinds of saponins was between 0. 6 ~ 4. 11 mg/g in the Panax notoginseng cream. The NIR quatitative model of three kinds of saponins was developed based on 6 102. 1 ~ 5 446. 4 cm^-1 as the predicted wavelength and four as dimension of maincomponents in Panax notoginseng cream. The correlation coefficient R2 of model was 0. 937 5 and calibration root mean square deviation( RMSEC) was0. 451; the prediction root mean square deviation( RMSEP) was 0. 588,the relative percent deviation( RPD) was 4. 69,and average prediction recovery was 103. 33% ± 11. 07%. The established model in this paper is accurate enough to determine the content of three saponinsin in Panax notoginseng cream. It can meets the requirements of rapid detection for the effective components of Panax notoginseng cream.
