摘 要:[目的]本研究通过基因表达汇编(gene expression omnibus,GEO)数据库探索核糖核苷酸还原酶M2(ribonucleotide reductase regulatory subunit M2,RRM2)在肺癌中的表达情况,探讨RRM2在肺癌发生发展过程中的作用。[方法]在美国国立生物技术信息中心(National Center for Biotechnology Information,NCBI)公共基因芯片数据库中检索并下载肺癌数据集,结合临床信息对芯片数据进行分析。利用基因集富集分析分析RRM2可能参与的信号通路以及生理过程。[结果]RRM2表达水平与性别、吸烟史、分化程度、AJCC T分期、AJCC N分期、病理类型相关(P〈0.05);同时,RRM2表达水平与肺癌患者的总体生存和复发相关(P〈0.05)。RRM2高表达的肿瘤样本富集了细胞周期调控、错配修复基因、氧化磷酸化、蛋白酶体、p53信号通路、Toll样受体的基因集。[结论]RRM2与肺癌患者的临床病理特征相关,可能为潜在的判断肺癌患者预后的指标及治疗肺癌的靶点。[Objective] To analyze the relationship between ribonucleotide reductase regulatory subunit Ma( RRM2) and clinicopathological characteristics of lung cancer by using existing databases. [Methods] The lung cancer datasets from Gene Expression Omnibus(GEO) of National Center for Biotechnology Information(NCBI) were searched and downloaded,the relationship between chip data and clinicopathological features of lung cancer was analyzed. Gene set enrichment analysis(GSEA) was conducted to explore the gene sets enriched in samples with high RRM2 expression. [Results] The higher expression of RRM2 was correlated with lower tumor differentiation,AJCC T stage and N stage and poorer prognosis of lung cancer. GSEA indicated that RRM2 regulating gene sets were associated with cell cycle,mismatch repair,oxidative phosphorylation,proteasome,p53 signaling pathway and Toll-like receptor pathway. [Conclusion] RRM2 might be used as an indicator for prognosis and as a therapeutic target for lung cancer patients.
