T-cadherin在胃癌组织中的表达及临床意义

摘　　要：目的:探讨胃癌组织中T-cadherin的表达及临床意义。方法:收集胃癌切除术后胃癌及癌旁组织166例,采用免疫组化、Western blot及Real-time PCR检测T-cadherin的蛋白及mRNA表达,并分析T-钙粘蛋白表达与临床病理特征及预后的关系。结果:与癌旁组织相比,T-cadherin的mRNA及蛋白表达均明显降低(P<0.05);胃癌组织中T-cadherin蛋白表达降低与吸烟、肿瘤体积较大(直径大于4cm)、淋巴结转移、分化差、临床分期高等因素相关(P<0.05或P<0.01),而与性别、年龄、饮酒和HP感染无明显相关性(P>0.05)。多因素分析表明,T-cadherin是胃癌患者生存的独立预后因素。结论:T-cadherin下调可能在胃癌的发生发展过程中起重要作用,T-cadherin可能是胃癌患者预后的标志物。Objective：The aim of this study was to evaluate the expression and significance of T-cadherin in gastric cancer.Methods：Gastric cancer and matched adjacent tissues from 166 patients receiving surgical resection were included.The expression of T-cadherin was detected by immunohistochemistry,Western blot and Real-time PCR.The expression of VEGF was detected by immunohistochemistry and its relationship with T-cadherin expression was analyzed.Meanwhile,association between T-cadherin expression and clinicopathologic feature data were analyzed.Results：The mRNA and protein expression of T-cadherin was significantly lower in gastric cancer tissues than the corresponding adjacent cancer tissues（P〈0.05）.Decreased expression of T-cadherin protein was correlated with smoking,larger tumor size（diameter4 cm）,lymph node metastasis,poor-differentiation and higher TNM stage（P〈0.05 or P0.01）.However,the expression of T-cadherin protein was not correlated with gender,age,acohol and Hp infection（P〈0.05）.The multivariate analysis showed that T-cadherin expression was an independent prognostic factor of the overall survival rate of patients with gastric cancer.Conclusions：The results indicate that Tcadherin is down-regulated in gastric cancer and may play an important role in the progression of gastric cancer.Tcadherin could be a valuable prognostic marker for gastric cancer patients.
