摘  要:目的观察血必净注射液对百草枯中毒大鼠肺组织中超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量及白细胞介素-6(IL-6)、转化生长因子-β_1(TGF-β_1)蛋白表达的影响,探讨血必净注射液治疗急性百草枯中毒的机制。方法采用随机数字表法将108只Wistar大鼠分成对照组、染毒组、干预组,每组36只。干预组造模后给予血必净注射液4 m L/kg腹腔注射,对照组、染毒组给予等量生理盐水腹腔注射。每组分别在干预后1,3,7,14,21,28 d各处死6只大鼠,检测肺组织中SOD活力、MDA含量及IL-6、TGF-β_1表达量。结果染毒组大鼠肺组织中SOD活力下降,MDA含量升高,IL-6和TGF-β_1蛋白表达增强,干预后各时间点与对照组比较差异均有统计学意义(P均<0.05)。干预组干预后各时间点SOD活力均明显高于染毒组(P均<0.05),但干预后1~14 d SOD活力均明显低于对照组(P均<0.05);干预后1~21 d MDA含量和各时间点IL-6、TGF-β_1表达量均明显低于染毒组(P均<0.05),但干预后1~14 d MDA含量和干预后各时间点IL-6、TGF-β_1表达量均明显高于对照组(P均<0.05)。结论氧自由基和细胞因子IL-6、TGF-β_1均参与百草枯所致肺损伤,而血必净注射液能够通过调节这些指标表达而减轻染毒大鼠肺组织损伤。Objective It is to observe the effects of Xuebijing injection on the activity of SOD,the content of MDA and the expression of IL-6,TGF-β1 in the lung tissue of paraquat-poisoned rats,thus to explore the mechanism of Xuebijing in the treatment of acute paraquat poisoning. Methods On the base of random number table,108 Wister rats were divided into3 groups: control group,poisoned group,treatment group,each group had 36 rats. After model establishing successfully the treatment group was given Xuebingjing injection 4 m L/kg by intraperitoneal injection,the other two groups were given the same dose of normal water. On 1 st,3 rd,7 th,14 th,21 st and 28 th day,six rats from each group were executed to detect the activity of SOD,the content of MDA and the expression of IL-6,TGF-β1 in the lung tissue. Results In the poisoned group,the activity of SOD in the lung tissue of the rats was decreased,the content of MDA was increased,and the expression of IL-6,TGF-β1 was increased,and there was a significant difference compared with the control group at every time point( P〈0. 05). After the treatment of Xuebijing,the SOD activity increased slightly,MDA content increased and IL-6 and TGF-β1 expression decreased in the treatment group,which was statistically significant compared with the poisoned group( P〈0. 05). Compared with the control group,the activity of SOD( 1 d to 14 d) was still lower( P〈0. 05),the content of MDA( 1 d to 14 d) and the expression of IL,TGF-β1 were still higher( P〈0. 05) in the treatment group. Conclusion Oxygen free radicals and cytokine IL-6,TGF-β1 play an important role in lung injury induced by paraquat in rats;Xuebijing can relieve the lung tissue injury of paraquat-poisoned rats by regulating these indexes.
