摘 要:目的探讨建立大鼠半乳糖性白内障模型的方法。方法 48只大鼠,随机分为8组,每组6只,双眼均为观察眼,采用腹腔注射法建立大鼠半乳糖性白内障。每日对每只大鼠进行称量,A组:50%D-半乳糖10 g/(kg·d);B组:50%D-半乳糖15 g/(kg·d);C组:50%D-半乳糖10 g/kg,每日2次;D组:50%D-半乳糖15 g/kg,每日2次;E、F、G、H的注射剂量等同对应A、B、C、D组,A、B、C、D组在第10日开始加喝10%D-半乳糖水。每3 d用裂隙灯观察晶状体的浑浊程度一次。结果观察至第30日时,除最低剂量的E组(33.33%)外,各实验组的造模成功率均为100%,A^H各组的死亡率(%)分别为0、33.33、66.67、33.33、0、16.67、0、16.67。总死亡率为20.83%,造模成功率为70.83%。结论腹腔注射半乳糖可以成功建立白内障动物模型,且造模的成功率较高,死亡率较低,是较稳定的半乳糖白内障动物模型。Objective To explore the method of establishment of rat models with galactose cataract. Methods Forty-eight rats were randomly divided into 8 groups( n = 6 ) . Bilateral eyes were regarded as the observation eyes. The rat models with galactose cataract were established by intraperitoneal injection. Each rat was weighed daily. In group A, 50% D-galactose 10 g/kg was administered once daily. In group B, 50% D-galactose 15 g/kg was given once daily. In group C, 50% D-galactose 10 g/kg was administered twice daily. In group D, 50% D-galactose 15 g/kg was injected twice a day. The injection doses in groups E, F, G and H corresponded to those in groups A, B, C and D, respectively. In groups A, B, C and D, 10% D-galactose water was supplemented on the 10 th day. The opacity of lenses was observed by the slit-lamp biomicroscope every 3 d. Results On the 30 th day, the success rate of each group was 100%, except for the group E (33.33%) in which the lowest dose was administered. The mortality rates in groups A-H were 0.00%,33.33%,66.67%,33.33%,0.00%,16.67%,0.00%,16.67%, respectively. The total mortality rate was 20.83%, and the success rate of model establishment was 70.83%. Conclusion Intraperitoneal injection of galactose can successfully establish the animal models of cataract with high success rate and low mortality rate. The established rat model with galactose cataract is relatively stable.
