摘 要:目的观察人类Apo E3基因过表达对大鼠肾上腺嗜铬细胞瘤细胞PC12凋亡的影响。方法将PC12转染含人Apo E3基因的p EGFP质粒及空载体p EGFP质粒,经持续筛选建立起稳定表达人Apo E3基因和质粒p EGFP的PC12的细胞系。将细胞分成4组:正常对照组(用正常培养基培养)、H_2O_2组(用400μmol/L H_2O_2诱导4 h)、H_2O_2+空质粒组(细胞转染p EGFP经筛选后用H_2O_2诱导4 h)、H_2O_2+Apo E3组(细胞转染Apo E3经筛选后用H_2O_2诱导4 h)。通过CCK-8检测细胞存活率,Hochest33258检测细胞凋亡。结果与正常对照组比较,H_2O_2组、H_2O_2+空质粒组和H_2O_2+Apo E3组细胞存活率下降(P<0.05),细胞凋亡增加。H_2O_2组和H_2O_2+空质粒组各指标比较,差异无统计学意义(P>0.05);H_2O_2+Apo E3组细胞存活率明显高于H_2O_2组、H_2O_2+空质粒组(P<0.05),H_2O_2组+Apo E3组细胞凋亡较H_2O_2组和H_2O_2+空质粒组明显减少。结论 Apo E3基因高表达对H_2O_2所致的PC12细胞氧化损伤有很好的保护作用。Objective To observe the effect of human Apo E3 gene high expression on the apoptosis of rat adrenal pheochromocytoma PC12 cells. Methods The p EGFP plasmid containing human Apo E3 gene and p EGFP plasmid were transfected with PC12 cells, and PC12 cell line was established that could stably express human Apo E3 gene and p EGFP plasmid through continuous screening. PC12 cells were allocated into 4 groups: normal control group, H2O2 group(cultured with 400 μmol/L H2O2 for 4 hours), liposome group(p EGFP transfection and H2O2 culture) and Apo E3 group(Apo E3 transfection and H2O2 culture). Cell survival was tested by CCK-8 method, cell apoptosis was detected by Hochest33258. Results Compared with the normal control group, cell survival rate was decreased in the other three groups(P〈0.05), while cell apoptotic was increased. The differences in the indexes mentioned above were not significant between H2O2 group and liposome group(P〈0.05). The cell survival rate was higher in the Apo E3 group, compared with H2O2 group and H2O2+liposome group(P〈0.05); the apoptosis was obviously lower in the Apo E3 group,compared with the H2O2 group and H2O2+liposome group. Conclusion The overexpression of Apo E3 gene has a great protective effect on the oxidative damage of PC12 cell mediated by H2O2.
