摘 要:对金针菇(Flammulina velutipes)菌种在木屑培养基上进行连续6次的继代培养,比较不同继代菌种与原始母种的菌丝生长速度、液体发酵pH以及工厂化栽培子实体的相关农艺性状的变化,利用基因组重测序技术检测继代菌种在DNA分子水平的遗传变异。结果显示:与原始母种相比,各继代菌种的表型性状无显著性差异,6次传代菌种的表型性状均稳定。DNA分子水平的遗传分析表明,第1~5次继代菌种基因组中的遗传变异呈缓慢增长趋势。与原始母种相比,第5次继代菌种的遗传差异达到7.92%,其它继代菌种的遗传差异均低于4%。采用木屑培养基进行金针菇菌种的继代培养可有效保持菌种的稳定性。The stability of Flammulina velutipes spawn subcultured on a sawdust medium was studied. The original strain was subcultured for six consecutive passages and then each subculture was compared to the original starting strain in terms of mycelial growth rate, pH in liquid fermentation, agronomic traits of fruiting body pertaining to the factory production of F. velutipes, and genetic deviations at the DNA level. Compared to the original strain, all the six passages of strains maintained high stability in phenotypic traits, and genetic variations for the first five passages gradually increased as the number of subcultures increased. Compared to the original strain, genetic differences of the first four subcultures remained less than 4%, and the genetic difference of the fifth subculture was 7.92%. This experiment showed that the spawn quality of F. velutipes was effectively maintained on the sawdust medium.
