摘 要:应用扩增片段长度多态性(AFLP)技术对以岱衢洋野生大黄鱼为亲本、通过群体选育方法获得的"东海1号"大黄鱼选育系F4代进行了遗传多样性分析.结果显示:16对AFLP选择性扩增引物组合在选育F4代的20尾个体中共扩增出736个位点,其中多态位点292个,多态率为39.67%;各引物对的Nei’s遗传多样性指数范围为0.090 4~0.203 7,平均值为0.139 9;Shannon多样性范围为0.132 0~0.304 7,平均值为0.209 9;其遗传多样性水平与原始亲本群体相当.表明若群体选育方法得当,多代选育后仍能维持较好的遗传多样性水平.Amplified fragment length polymorphism(AFLP) technique was used to analyze the genetic diversities of the 4 th generation(F4) of "Donghai No.1" large yellow croaker(Larimichthys crocea) breeding line. The breeding line was derived from the wild large yellow croaker that captured from Daiquyang in Zhejiang Province. Using 16 AFLP selective primer-pairs, a total of 736 loci were detected from 20 individuals, of which 292 loci were polymorphic. The percentage of polymorphism was 39.67%. The range of Nei's genetic diversity and Shannon genetic diversity index were 0.090 4-0.203 7(mean 0.139 9) and 0.132 0-0.304 7(mean 0.209 9), respectively. The genetic diversity of F4 was at the similar level compared to original parents. It indicates that the genetic diversity would not decrease significantly after several generations with proper mass selection method.
