摘  要:目的:探讨一种尾静脉注射人源肺癌循环肿瘤细胞建立的转移模型,比较NOD/SCID小鼠、Nude小鼠和C57BL/6小鼠对该模型的敏感性。方法:将人源肺癌循环肿瘤细胞通过尾静脉注射接种到C57BL/6小鼠、Nude小鼠和NOD/SCID小鼠体内,在不同时间节点对不同品系小鼠上的肺转移情况进行评估;H&E染色检测肺组织病理变化。结果:CTC-TJH-01细胞接种于NOD/SCID小鼠体内后,在第8周首次观察到肺部成瘤,在第10周小鼠肺部成瘤率为100%。而在Nude小鼠和C57BL/6小鼠体内均不成瘤。结论:人源肺癌循环肿瘤细胞通过尾静脉注射接种于NOD/SCID小鼠后能够发生肺转移,该模型可用于临床抗转移药物筛选与评价研究。Objective: To investigate a metastasis model established by tail vein injection of human lung cancer circulating tumor cells,and compare the sensitivity of NOD/SCID mice,nude mice and C57 BL/6 mice to this model. Methods: Human lung cancer circulating tumor cells were inoculated into C57 BL/6 mice,Nude mice,and NOD/SCID mice via tail vein injection. The lung metastases of different strains of mice were assessed at different time points; HE staining was used to detect the pathological changes of lung tissue. Results:After CTC-TJH-01 cells were inoculated into NOD/SCID mice,the lung tumors were observed for the first time at the 8 th week. In the 10 th week,the rate of tumor formation in the lungs of mice was 100%. However,tumors were not formed in Nude mice and C57 BL/6 mice. Conclusion: Human lung cancer circulating tumor cells can invade lung metastases after inoculation in NOD/SCID mice by tail vein injection.This model can be used for the screening and evaluation of clinical anti-metastatic drugs.
