摘  要:目的探讨常见肿瘤显像剂2-[^(18)F]-2-脱氧-β-D-葡萄糖([^(18)F]-FDG)在西门子Explora One化学合成模块上全自动合成方法、质量控制以及影响因素。方法西门子Eclipse RD回旋加速器生产出[^(18)F-]与以三氟甘露糖为前体进行亲核取代反应合成乙酰化[^(18)F]-FDG,高纯氮气推动,Sep-Pak~C_(18)萃取柱吸附中间体,氢氧化钠水解C-18柱上中间产物,经纯化,灭菌最终得到[^(18)F]-FDG注射液。通过薄层色谱法对合成的产品进行放射化学纯度(RCP)检测,肿瘤患者行[^(18)F]-FDG PET-CT扫描。结果最终合成产物[^(18)F]-FDG的RCP>95%,20 mg的三氟甘露糖可获得未校正合成效率约为60%。结论使用ABX[^(18)F]-FDG套件,Explora One化学合成模块实现固相萃取法合成PET显像剂[^(18)F]-FDG,合成时间短,稳定快捷。Objective To present a method for automatic synthesis of common tumor imaging agents [^18F]-FDG by Explora One module from Siemens corporation,and discuss the method for quality control,factors that affect synthesis of the tracers. Methods Siemens Eclipse RD cyclotron firstly produced [^18F-]and interacted with a precursor( 1,3,4,6-4-O-acetyl-2-O-trifluorosulfonyl-beta-Dpyranose) of [^18F]-FDG. Acetylated-[^18F]-FDG was pushed with high purity nitrogen and the intermediates( acetylated-[^18F]-FDG)captured by Sep-Pak~C18 extraction column. Na OH hydrolysis the intermediate carried by C-18 column,after purification,sterilization and eventually get [^18F]-FDG injection. The radiochemical purity( RCP) of the synthesized products was detected by TLC. PETCT scan of [^18F]-FDG tumor patient was carried out and confirmed by pathology. Results The RCP of [^18F]-FDG is over 95%. 20 mg of trifluorocarbon can achieve an uncorrected synthetic efficiency of about 60%. Conclusions It is easily and steadily to automatically synthesis the tumor imaging agents [^18F]-FDG by using ABX [^18F]-FDG cassette with Explora One module. The synthesis time was short,stable and fast.
