摘 要:目的研究非小细胞肺癌组织中PTEN和mTOR的表达及意义。方法采用免疫组化方法检测非小细胞肺癌组织105例和癌旁组织60例中PTEN和mTOR的表达情况及与临床病理的关系。结果 mTOR蛋白在肺癌组织中的表达明显高于癌旁组织(75.2%vs 16.7%),PTEN蛋白在肺癌组织中的表达低于癌旁组织(48.6%vs 95.0%),差异有统计学意义(P<0.05)。PTEN蛋白在肺癌组织中的表达与分化程度、TNM分期和淋巴结转移情况有关(P<0.05),而与性别、年龄、组织学分型和肿瘤直径无关(P>0.05);mTOR蛋白在肺癌组织中的表达与淋巴结转移情况有关(P<0.05),而与性别、年龄、分化程度、组织学分型、TNM分期和肿瘤直径无关(P>0.05)。相关性分析显示:mTOR在非小细胞肺癌中表达与PTEN表达呈负相关(r=-0.583,P=0.001)。结论非小细胞肺癌组织中PTEN蛋白缺失可能与肺癌的发生与发展有关。Objective To investigate the expression and implication of PTEN and mTOR in human non-small cell lung cancer.Methods Expression of PTEN and mTOR was detected with immunohistochemistry SP assay in 105 cases of non-small cell lung cancer tissues and 60 cases of adjacent non-cancerous tissues, and its relationship with clinical pathological features was analyzed.Results The positive expression of mTOR proteins in NSCLC tissues was significantly higher than that in adjacent non-cancerous tissues, while the positive expression of PTEN proteins in NSCLC tissues was significantly lower than that in adjacent non-cancerous tissues( P 〈0.05).PTEN positive expression was significantly different among patients with different tumor differentiation, lymph node metastasis or TNM stages( P 〈0.05), but there was no significant difference among patients with different sex, age, histological classification and tumor size( P 〉0.05).The mTOR positive expression was significantly different in different lymph node metastasis( P 〈0.05), but there was no significant difference among patients with different sex, age, histological classification, tumor differentiation, TNM stages and tumor size( P 〉0.05).The expression of mTOR was negatively correlated with PTEN expression in NSCLC( r =-0.583, P =0.001).Conclusion The results suggest that the loss of PTEN expression in NSCLC may promote the occurrence and development of lung cancer.
