摘  要:目的探讨microRNA-128-3p(miR-128-3p)对人脐静脉内皮细胞(HUVEC)体外血管管腔形成能力的影响,并初步阐明其抑制血管生成的机制。方法利用慢病毒表达载体转染技术建立miR-128-3p稳定高表达的HUVEC细胞株,采用实时定量PCR检测miR-128-3p的表达,western blot检测miR-128-3p对HUVEC血管内皮生长因子家族(VEGF)及其受体(VEGFR)的表达水平,Matrigel胶体外管腔形成实验检测miR-128-3p表达对HUVEC的血管生成能力的影响。结果 qRT-PCR显示本研究成功建立了慢病毒载体介导的miR-128-3p过表达的HUVEC细胞株,western blot检测发现体外过表达miR-128-3p可明显抑制HUVEC中VEGF家族因子VEGF-A、VEGF-D及其受体VEGFR-2、VEGFR-3的表达(P<0.05),体外管腔形成实验显示过表达miR-128-3p的HUVEC的体外管腔形成数量明显低于对照组的管腔形成数量,具有统计学意义(P<0.05)。结论 miR-128-3p可通过抑制VEGF家族因子及其受体的表达抑制HUVEC管腔形成能力。Objective To investigate the effect of microRNA-128-3 p( miR-128-3 p) on human umbilical vein endothelial cells( HUVEC) tube formation in vitro and explore its possible mechanism. Methods Lentivirus vector infection technology was used to specifically induce the expression of miR-128-3 p in HUVEC. Quantitative real-time polymerase chain reaction( qRT-PCR)was used to detect the expression level of miR-128-3 p. Western blot was used to detect the expression level of vascular endotheilal grow factor( VEGF) and vascular endotheilal grow factor receptor( VEGFR) in HUVEC. Matrigel-plug assay was used to detect the tube formation ability of HUVEC in vitro. Results QRT-PCR showed that miR-128-3 p over-expression HUVEC cell line was successfully established by lentivirus vector infection technology. Western blot assay showed that miR-128-3 p over-expression inhibit the expression level of VEGF-A,VEGF-D,VEGFR-2 and VEGFR-3 in HUVEC( P 〈 0. 05). Matrigel-plugassay showed that the tube formation numbers in miR-128-3 p over-expression HUVEC cell line was significantly decreased than that in control group( P 〈 0. 05). Conclusion miR-128-3 p can inhibit HUVEC tube formation by reduce the expression of VEGF and VEGFR.
