摘 要:利用固相萃取小柱,建立杉木种子酚类提取液的纯化工艺,并优化固相萃取条件.结果表明:采用Bond Elut Plexa固相萃取小柱,确定淋洗液为体积分数为35%的甲醇,洗脱液为80%甲醇,有效去除和分离了种子提取液中的大量干扰成分,在此条件下总酚回收率达到70%以上.以不同家系授粉后100 d杉木种子为材料验证优化工艺,测定9种酚类化合物含量,L、H、M家系含量最大分别为莽草酸、对羟基苯甲酸和绿原酸,最低则为没食子酸、芦丁和丁香酸.研究结果不仅为后期监测杉木种子发育过程中的酚类物质的动态变化奠定基础,并且为今后研究杉木种子的生理生化特征提供一定的技术和理论支持.The purification process of polyphenol seed extract of Chinese fir was established by solid phase extraction column and the conditions of solid phase extraction were optimized. The results showed that Bond Elut Plexa solid-phase extraction column, the dection condition was determined to be 35% methanol as an elution liquid and 80% methanol as a washing liquid.Under the condition, a large number of interfering components in the seed extract were effectively separated and the recovery rate of polyphenols was over 70%. The optimized process was verified by pollination of different varieties 100 d fir seeds. The content of 9 kinds of phenolic compounds was determined. The largest content of L, H and M families were shikimic acid, 4-Hydroxybenzoic acid and chlorogenic acid while the lowest was Gallic acid, rutin and Syringic acid. The results not only provide a basis for monitoring the dynamic changes of phenolic substances in the development of Chinese Fir Seeds, but also provide some technical and theoretical support for the study of the physiological and biochemical characteristics of Chinese fir seeds in the future.
