摘 要:目的研究阿司匹林(Aspirin)对小鼠胶质瘤干细胞增殖及凋亡的影响。方法体外培养小鼠胶质瘤GL261细胞株的干细胞,分为对照组(加入等体积生理盐水)和阿司匹林2.5、5、10 mmol/L处理组(n=5)。应用CCK-8试剂盒检测细胞增殖情况,流式细胞仪检测细胞周期,TUNEL染色检测细胞凋亡情况,LDH试剂盒检测细胞受损情况。结果与对照组相比较:不同浓度的阿司匹林均可抑制胶质瘤干细胞的增殖(P<0.05),并且呈剂量—时间依赖性。各种浓度的阿司匹林作用后胶质瘤干细胞的成球能力明显下降(P<0.05)。阿司匹林影响胶质瘤干细胞的细胞周期,S期的细胞比例减少,G0/G1期的细胞比例增加(P<0.05)。3种浓度阿司匹林作用3 d后,胶质瘤干细胞的凋亡细胞数目增多(P<0.05),细胞释放LDH明显上升(P<0.05)。结论阿司匹林具有抑制胶质瘤干细胞的增殖,并促进其凋亡的作用。Objective To study the effects of aspirin on proliferation and apoptosis of mouse glioma stem cells( GSCs). Methods In vitro cultured GSCs isolated from mouse glioma cell line GL261 were treated with 2. 5,5. 0 and 10 mmol/L aspirin,with normal saline as the control. The proliferation of GSCs was assayed using CCK-8 kit,and the changes in cell cycle distribution and cell apoptosis were detected with flow cytometry and TUNEL assay,respectively; LDH kit was used to assess the cell damage after aspirin treatment. Results Compared with the control cells,the GSCs treated with different concentrations of aspirin showed dose-and time-dependent inhibition of cell proliferation( P〈0. 05) and obviously reduced number and size of the tumor spheres( P〈0. 05). Aspirin caused obvious cell cycle changes in GSCs,shown by a significantly decreased percentage of S phase cells and an increased percentage of G0/G1 phase cells( P〈0. 05). Aspirin also induced obvious apoptosis of GSCs and significantly increased the release of cytosolic LDH( P〈0.05). Conclusion Aspirin inhibits the proliferation and promotes apoptosis of mouse GSCs in vitro.
