摘 要:本文利用DEAE-52纤维素层析柱、Sephadex-G200和Sephadex-G150层析柱对羧甲基茯苓粗多糖进行分离纯化,得到一种纯化多糖组分,不仅鉴定了其结构,而且也探究了其体外抗肿瘤活性和抗炎活性。该多糖是一种分子量为20.96×104 u的(1→3)-β-D-葡聚糖,含有少量(1→6)和(1→2)糖苷键,单糖组成为只包含D-葡萄糖,具有三螺旋结构。该多糖对人结肠癌细胞HT-29、人肝癌细胞Hep G-2、人胃癌细胞SGC-7901、人乳腺癌细胞MCF-7以及人肺癌细胞A549的细胞增殖具有抑制作用,其IC50值分别为140.5μg/m L、264.3μg/m L、102.5μg/m L、256.4μg/m L、313.2μg/m L。此外,该多糖对脂多糖(LPS)诱导的小鼠巨噬细胞RAW264.7有一定的抑制作用,而在无LPS刺激的情况下对该细胞反而具有一定的激活效果。研究结果为以茯苓多糖为主要活性成分的保健食品和药品开发提供了理论基础和技术支持。A carboxymethyl polysaccharide from Pachyman was isolated and purified using DEAE-52 cellulose column, Sephadex-G200 column and Sephadex-G150 column in the work. The structure of this polysacchairde was identified and its anti-proliferation activity and anti-inflammation activity investigated. This polysaccharide is a(1→3)-β-D-glucan with an average molar mass of 20.96×104 u, which also has a little(1→2) glucosidic bonds. Its monosaccharide composition only contained D-glucose and the triple helical structure did not exist. This polysacchairde exhibited a strong anti-proliferation effect on HT-29, Hep G-2, SGC-7901, MCF-7 and A549 cells, with IC50 values of 140.5 μg/m L, 264.3 μg/m L, 102.5 μg/m L, 256.4 μg/m L and 313.2 μg/m L, respectively. In addition, this polysaccharide exhibited an inhibitory effect on LPS-induced RAW264.7 cells, but could activate RAW264.7 cells in the absence of LPS. The findings would be of great importance for the development of Pachyman polysaccharide-based health foods and drugs.
