摘　　要：目的探讨Y-box结合蛋白1(YB-1)在结肠癌组织中的表达情况,并分析与临床病理特征的相关性。方法分别运用Real-time Quantitative PCR及免疫组织化学染色方法检测YB-1在结肠癌组织中的表达情况。结果 YB-1在结肠癌组织中的表达显著高于配对的正常组织;在90例正常组织中,YB-1表达阴性52例(57.78%),弱阳性27例(30.00%),强阳性11例(12.22%);90例结肠癌组织中,YB-1表达阴性7例(7.78%),弱阳性34例(37.78%),强阳性49例(54.44%);二者差异有统计学意义(P<0.05)。YB-1在结肠癌组织中的表达水平与临床AJCC分期、肿瘤进展程度(T)、淋巴结转移(N)、远处转移(M)、组织学分型及脉管浸润等临床病理特征相关(P<0.001),而与年龄、性别、肿瘤位置等临床病理特征无关。结论 YB-1在结肠癌中异常高表达,其表达水平与肿瘤的恶性生物学行为显著相关,可能是新型结肠癌肿瘤标志物及治疗靶点。Objective To study the expression of Y box binding protein 1（ YB-1） and evaluate whether YB-1 was associated with histopathological features in the patients with colon cancer or not. Methods Real-time Quantitative PCR and immunohistochemistry was performed to analyze the association between YB-1 expression and clinical pathological features on a tissue micro array containing 90 cases of colon cancer. Results The expression of YB-1 was overexpressed in colon cancer compared with normal mucosa. Immunohistochemical study displayed that YB-1 expression was with negative（ 57. 78%）,weak positive（ 30%）,strong positive（ 12. 22%） in normal mucosa and YB-1 expression was with negative（ 7. 78%）,weak positive（ 37. 78%） and strong positive（ 54. 44%） in colon cancer tissues. Overexpression of YB-1 was significantly associated with the AJCC staging,depth of tumor invasion,lymph node metastasis,distant metastasis,histological differentiation and vascular invasion. Conclusion The high expression of YB-1 is closely correlated with the malignant biological behavior of colon cancer,which can be served as a novel prognostic marker and therapy target of colon cancer.
