摘 要:目的优选芩龙降压胶囊的提取工艺。方法以龙胆苦苷、黄芩苷、芍药苷及毛蕊花糖苷含量的综合评分为指标,采用L_9(3~4)正交试验法考察加水量、提取次数及提取时间对芩龙降压颗粒水提工艺的影响。采用RP-HPLC测定龙胆苦苷、芍药苷含量,流动相为乙腈-0.1%磷酸水溶液(9:91);采用RP-HPLC测定黄芩苷含量,流动相为乙腈-0.1%磷酸水溶液(21:79);采用RP-HPLC测定毛蕊花糖苷含量,流动相为乙腈-0.1%磷酸水溶液(16:84);流速1.0 ml/min,检测波长依次为270,230,280和334 nm。结果最佳提取工艺为加8倍量水提取3次,每次2 h;黄芩苷、龙胆苦苷、芍药苷、毛蕊花糖苷质量分数分别为12.88%,10.03%,1.55%和0.78%。结论优选的提取工艺稳定可行,适用于该制剂的工业生产。Objective To optimize the extraction technology of Qinlong Jiangya Capsules. Methods With the comprehensive scores of the contents of baicalin, gentiopicroside, paeoniflorin and mulberry glycoside as indicators, L9(3-4) orthogonal test was adopted to optimize the extraction process by taking the amount of water, extraction frequency and time as factors. The contents of gentiopicroside, paeoniflorin, baicalin and mulberry glycoside were determined by RP-HPLC, and the mobile phases were acetonitrile-0.1 % phosphoric acid mixures at the ratios of 9:91, 9:91, 21:79 and 16:84, respectively. The flow rate was 1.0 ml/min. The detection wavelengths were 270, 230, 280 and 334 nm, respectively. Results The optimum extraction process was as follows: the medicinal materials were extracted by reflux for 3 times with 8-fold water, 2 h for each time. The mass fractions of baicalin, gentiopicroside, paeoniflorin and mulberry glycoside were 12.88 %, 10.03 %, 1.55 % and 0.78 %, respectively. Conclusion The optimized extraction technology can be stable, feasible and suitable for the industrial production of Qinlong Jiangya Capsules.
