血清microRNA-19a在非小细胞肺癌中的表达及其临床意义

摘　　要：目的探讨血清microRNA-19a(miR-19a)在非小细胞肺癌(NSCLC)中的表达及其临床意义。方法采用荧光定量PCR方法检测50例NSCLC患者和60例健康对照者血清miR-19a表达,同时测定患者及对照者血清癌胚抗原(CEA)、细胞角蛋白片段19(CYFRA21-1)、神经元特异性烯醇化酶(NSE)的表达水平。结果 NSCLC患者血清miR-19a相对表达量(3.46±1.33)显著高于对照组(1.10±0.48),且TNMⅢ~Ⅳ期患者miR-19a相对表达量(4.00±0.99)明显高于TNMⅠ~Ⅱ期患者(3.03±1.41),差异均有统计学意义(P<0.05)。ROC曲线分析显示,miR-19a、CEA、CYFRA21-1及NSE的曲线下面积(AUC)分别为0.910、0.752、0.856及0.817。结论 miR-19a也许可作为一种新的标志物用于NSCLC的早期诊断。Objective To investigate the expression level and clinical significance of microRNA-19 a（miR-19 a）in the serum of patients with non-small cell lung cancer（NSCLC）.Methods Serum level of miR-19 awas measured by Quantitative real-time PCR,and the levels of carcinoma embryonic antigen（CEA）,cytokeratin protein fragment 21-1（CYFRA21-1）,and neuron specific enolase（NSE）were also measured at same time in50 patients with NSCLC and 60 healthy control subjects.Results The relative expression level of miR-19 a was significantly increased in patients with NSCLC（3.46±1.33）compared with healthy control subjects（1.10±0.48）,and the patients at TNM stages Ⅲ-Ⅳ（4.00±0.99）had higher miR-19 aexpression than those at TNM stagesⅠ-Ⅱ（3.03±1.41）,and the differences were statistically significant（P〈0.05）.The areas under receiver operating characteristic curve（AUC）of serum miR-19 ain diagnosis of NSCLC was0.910,and the AUC of CEA,CYFRA21-1 and NSE was 0.752,0.856 and 0.817,respectively.ConclusionThe serum miR-19 amay be a novel marker for early diagnosis of NSCLC.
