摘 要:目的探究PRMT7在肺癌转移中的功能,并找到调控PRMT7的信号分子。方法通过免疫蛋白共沉淀法得到一系列与PRMT7相互作用的蛋白,通过蛋白质谱分析,结合生物信息学的方法,筛选具有显著意义的相互作用蛋白,并进行验证。结果确认了HSPA5等5个候选基因,经过对各种肺癌数据库的分析发现,候选基因的表达水平高于其他基因。结论 PRMT7与HSPA5存在相互作用,此发现有助于阐明PRMT7对肺癌细胞入侵和转移的促进作用。Objective To explore the role of PRMT7 in lung cancer metastasis and find out the signaling molecule which can regulate PRMT7. Methods A series of proteins in interaction with PRMT7 were obtained by coimmu- noprecipitation,and proteins of interaction with significance were screened out and tested by protein mass spectrometry and bioinformatics. Results Five candidate genes were identified. The analysis of various databases about lung cancer discovered that the expression level of candidate gene was higher than that of other genes. Conclusion PRMT7 can interact with HSPA5,which is helpful to clarify the promotion function of PRMT7 in the invasion and metastasis of lung cancer cell.
