摘 要:目的通过给高糖高脂喂养的Wistar大鼠腹腔注射链脲佐菌素(streptozotocin,STZ)结合牙间结扎正畸钢丝,建立一种近似人糖尿病牙周炎的大鼠模型,探索中药单体人参皂甙Rg1对糖尿病牙周炎模型大鼠的疗效及机制研究。方法选用100只体质量160~200 g的Wistar大鼠,按照随机数字表法分成10组,每组10只。A组:正常对照组,0周时处死。B-J组建立糖尿病牙周炎大鼠模型。B-D组:人参皂甙Rg-1治疗组,第1、2、3周处死。E-G组:甲硝唑治疗组,第1、2、3周处死。H-J组:糖尿病牙周炎对照组,第0、1、2周处死。取大鼠上颌磨牙牙周组织标本作切片,光镜观察,免疫组化方法对IL-1β、IL-6、PGE-2进行染色,测量并分析灰度值。结果 B-D组和E-G组牙周组织中IL-1β、IL-6、PGE-2水平均显著低于H-J组(P<0.05),同时B-J组各指标显著高于A组(P<0.05)。结论人参皂甙Rg1可降低糖尿病牙周炎大鼠牙周组织中的炎性细胞因子水平,改善大鼠的血糖状态,减轻牙周组织的炎症程度并改善其全身状况。Objective To establish an animal model of human clinical diabetic periodontitis in order to study the effect and mechanism of ginsenoside Rg1 in the treatment of diabetic periodontitis in rats. Methods One hundred Wistar rats that weighed 160 to 200 g were selected and equally divided into 10 groups using the random number table method. Group A: the normal control group that was executed at 0 week. A rat model of diabetic periodontitis was established in Groups B to J. Groups B to D: ginsenoside Rg-1 treatment group,where rats were sacrificed at the first,second,third weeks respectively. Groups E to G:metronidazole treatment group,where rats were sacrificed at the first,second,third weeks respectively. Groups I-J: diabetic periodontitis control group,where rats were executed at 0,first and second weeks. The specimens of rat periodontal tissues from maxillary molars were sectioned and observed with light microscopy. Immunohistochemical staining was used to stain IL-1,IL-6 and PGE-2,and the gray values were measured and analyzed. Results The levels of IL-1 beta,IL-6 and PGE-2 in periodontal tissues of Groups B to D and Groups E to G were significantly lower than those of Groups I to J( P〈0.05),while indexes of Groups B to J were significantly higher than those of Group A( P〈0.05). Conclusion Ginsenoside Rg1 can reduce the level of inflammatory cytokines in rats with periodontitis,improve blood glucose in rats,reduce the degree of inflammation in periodontitis and improve the conditions of the whole body,which provides evidence for the role of ginsenoside Rg-1 in the treatment of diabetic periodontitis.
