摘 要:目的:在Transwell细胞跨膜培养系统中培养T84细胞单层模拟肠黏膜屏障,并对Th17/Treg细胞相关因子处理前后T84细胞单层的跨上皮电阻(TER)和辣根过氧化物酶(HRP)通过率进行检测,以探讨其对肠黏膜屏障通透性的影响。方法:将不同浓度的IL-17、IL-22和IL-10因子分别加入到Transwell跨膜系统培养的T84细胞单层中,之后测量不同时间(0、6、12、24和48 h)T84细胞单层的TER和不同时间(6、24和48 h)HRP通过率的变化。结果:当不同浓度IL-17和IL-22处理时间大于6 h时,继续增加细胞因子的浓度和继续延长作用时间,T84细胞单层TER会逐渐下降并且HRP通过率逐渐增加。当IL-17和IL-22浓度大于100 ng/ml,继续增加其浓度,T84细胞单层的TER和HRP通过率均未见明显变化。当不同浓度IL-10作用于T84细胞后,随着时间和浓度的变化,T84细胞单层TER和HRP通过率均未发生明显变化。结论:在一定的浓度范围内,IL-17和IL-22均可以造成T84细胞单层通透性的增加,并且随着浓度的增加和作用时间的延长,T84细胞单层通透性的增加也越来越明显。但是IL-10并不影响T84细胞单层的通透性。Objective: The culture of T84 cell monolayers in the Transwell cell transmembrane culture system can be used to simulate the intestinal mucosal barrier. To explore its effect on the permeability of intestinal mucosal barrier,we added Th17/Treg cell related factors to the T84 cell monolayer,and detected the passing rate of the cross epithelial resistance( TER) and horseradish peroxidase( HRP) before and after its treatment. Methods: We added different concentrations of IL-17,IL-22 and IL-10 to the T84 cell monolayer culture of Transwell transmembrane system respectively. After that,the TER of T84 cell monolayers at different times( 0,6,12,24,and 48 h) and HRP passing rates at different times( 6,24 and 48 h) were measured. Results: When the concentration of IL-17 and IL-22 treatment time is greater than 6 h,with the extension of time and increase of cytokine concentration,T84 monolayer TER gradually decreased and HRP passing rate increased gradually. When the concentration of IL-17 and IL-22 was greater than 100 ng/ml and continued to increase its concentration,the TER and HRP passing rate through the T84 cell monolayer was not significantly changed. When the IL-10 of different concentrations acted on T84 cells,with the change of time and concentration,the T84 cell monolayer TER and HRP passing rate did not change significantly. Conclusion: Within a certain range of concentration,both IL-17 and IL-22 can increase the permeability of T84 monolayer. Moreover,with the increase of concentration and the prolongation of the action time,the permeability of T84 cell monolayer is more obvious. However,IL-10 did not affect the permeability of T84 cell monolayers.
