辛伐他汀和普伐他汀对人舌鳞癌Tca-8113细胞的增殖、凋亡作用影响的初步探究

摘　　要：目的:初步探究辛伐他汀和普伐他汀对人舌鳞癌Tca-8113细胞的促进增殖抑制及诱导细胞凋亡的作用。方法:采用不同浓度(15、30、45、60、75μmol/L)的辛伐他汀和普伐他汀分别作用于Tca-8113细胞不同的时间(24/48h),采用MTT法检测两种药物对Tca-8113细胞的体外增殖抑制作用;光学显微镜下观察Tca-8113细胞形态学变化;采用流式细胞仪检测细胞凋亡情况。结果:辛伐他汀和普伐他汀均可抑制Tca-8113细胞的增殖与促进凋亡,呈现浓度和时间依赖性(P<0.05)。结论:辛伐他汀和普伐他汀对人舌鳞癌细胞均有明显的抑制增殖、促进凋亡的作用,其中辛伐他汀的作用略高于普伐他汀。Objective：To study the effects of simvastatin and pravastatin on the proliferation and apoptosis of human tongue squamous cell carcinoma Tca-8113 cells.Methods：Tca-8113 cells were treated with 5 different concentrations,including 15,30,45,60,75μmol/L of simvastatin or pravastatin for 24 and 48 h.MTT assays was used to test the inhibition effect of simvastatin and pravastatin on Tca-8113 cells in vitro;optical microscope observe Tca-8113 cells morphology changes;annexin V-FITC/PI staining flow cytometry was used for cell apoptosis.Results：Simvastatin and pravastatin were all inhibited cells proliferation and induced apoptosis in dose-dependent and time-dependent manner（P〈0.05）.Conclusion：Simvastatin and pravastatin inhibit the cell growth of Tca-8113 cells,simvastatin shows a better effect than pravastatin on the cell proliferation and apoptosis in vitro.
