摘  要:目的探讨SDF-1/CXCR4在舌癌组织中的表达及对舌癌细胞系Tca8113增殖、迁移和侵袭的影响。方法 RTPCR及Western blot检测舌癌组织及癌旁组织中SDF-1和CXCR4的mRNA和蛋白表达;培养人舌癌细胞系Tca8113,将细胞分为对照组、SDF-1组、SDF-1+AMD3100组、AMD3100组,Western blot检测四组细胞CXCR4、MMP-2、MMP-9蛋白表达,CCK8试验检测四组细胞增殖,Transwell小室检测四组细胞迁移和侵袭数。结果舌癌组织中SDF-1和CXCR4的mRNA和蛋白表达均显著高于癌旁组织(P<0.01);SDF-1组CXCR4、MMP-2、MMP-9蛋白表达及细胞增殖、迁移、侵袭均显著高于另外三组(P<0.01);SDF-1+AMD3100组CXCR4蛋白表达显著低于对照组和SDF-1组;MMP-2、MMP-9蛋白表达及细胞增殖、迁移、侵袭与对照组差异不显著(P>0.05),但均显著低于SDF-1组;CXCR4、MMP-2、MMP-9蛋白表达及细胞增殖、迁移、侵袭均显著高于AMD3100组(P<0.01);AMD3100组CXCR4、MMP-2、MMP-9蛋白表达及细胞增殖、迁移、侵袭均显著低于另外三组(P<0.01)。结论 SDF-1和CXCR4在舌癌组织高表达,SDF-1可上调CXCR4在人舌癌细胞系Tca8113的表达,其过程能被AMD3100减弱。SDF-1/CXCR4可促进细胞的增殖、侵袭和迁移,此过程可被AMD3100抑制。Objective To investigate the expression of SDF-1/CXCR4 in tongue carcinoma and its effect on proliferation,migration and invasion of tongue carcinoma Tca8113 cell lines. Methods The mRNA and protein expressions of SDF-1 and CXCR4 in tongue carcinoma and paracancerous tissue were detected by RT-PCR and Western blot. Human tongue carcinoma Tca8113 cell lines were cultured,and cells were divided into the control group,SDF-1 group,SDF-1 + AMD3100 group and AMD3100 group. The expressions of CXCR4,MMP-2 and MMP-9 protein were detected by Western blot. The proliferation of cells in the four groups was detected by CCK8 test,while cell migration and invasion in the four groups were detected by Transwell. Results The mRNA and protein expressions of SDF-1 and CXCR4 in tongue carcinoma were significantly higher than those in paracancerous tissue( P〈0. 01). The expressions CXCR4 MMP-2,MMP-9 protein and proliferation,migration and invasion of cells in the SDF-1 group were significantly higher than those in the other three groups( P〈0. 01). The expression of CXCR4 protein in the SDF-1+AMD3100 group was significantly lower than that of the control group and SDF-1 group,while the expressions of MMP-2 and MMP-9 protein and cell proliferation,migration,invasion were not significantly different compared with the control group( P〈0. 05),but were significantly lower than in the SDF-1 group,and CXCR4,MMP-2,MMP-9 protein expressions and cell proliferation,migration and invasion were significantly higher than those of the AMD3100 group( P〈0. 01).The expressions of MMP-2,CXCR4,MMP-9 protein and proliferation,migration,invasion of cells in the AMD3100 group were significantly lower than those of the other three groups( P〈0. 01). Conclusion There are high expressions of SDF-1 and CXCR4 in tongue carcinoma,and SDF-1 can upregulate the expression of CXCR4 in human tongue carcinoma Tca8113 cell lines,but the process can be reduced by AMD3100.
