摘 要:目的探讨8q24rs6983267基因多态性与前列腺癌易感性的相关性。方法检索PubMed、Highwire、Cochrane library、Embase、维普、万方医学网、中国知网和中国生物医学文献数据库中所有关于8q24rs6983267基因多态性与前列腺癌易感性的文献,2位研究者根据纳入及排除标准独立对文献进行筛选、资料提取和质量评价后,对纳入数据进行Meta分析。结果共纳入相关文献16篇,包括21项研究,涉及13 925例病例和13 398例对照。Meta分析结果,超显性遗传模型(GG+TT vs GT:OR=1.02,95%CI:0.97~1.07,P=0.48)显示8q24rs6983267基因多态性与前列腺癌易感性无统计学相关;显性遗传模型(GG vs GT+TT:OR=1.26,95%CI:1.19~1.33,P<0.001)、隐性遗传模型(GG+GT vs TT:OR=1.26,95%CI:1.15~1.38,P<0.001)与纯合子对比模型(GG vs TT:OR=1.46,95%CI:1.31~1.62,P<0.001)显示8q24rs6983267基因多态性与前列腺癌易感性的相关性有统计学意义。结论 8q24rs6983267基因多态性与前列腺癌易感性呈正相关。Objective To explore the relationship between 8 q24 rs6983267 polymorphism and susceptibility to prostate cancer. Methods We obtained original research articles concerning the relationship between 8 q24 rs6983267 polymorphism and susceptibility to prostate cancer by searching PubMed and other databases.Two researchers independently screened the articles based on the inclusion and exclusion criteria.After the extraction and assessment of included data,the meta-analysis was applied to analyze included data. Results Sixteen relevant literatures including 21 studies were identified,containing 13 925 cases and 13 398 controls.The over-dominant model showed no significant association between 8 q24 rs6983267 polymorphism and susceptibility to prostate cancer(GG+TT vs GT:OR=1.02,95% CI:0.97-1.07,P=0.48).However,the dominant model(GG vs GT+TT:OR=1.26,95% CI:1.19-1.33,P〈0.001),recessive model(GG+GT vs TT:OR=1.26,95% CI:1.15-1.38,P〈0.001)and homozygote comparison(GG vs TT:OR=1.46,95% CI:1.31-1.62,P 〈0.001)showed statistical significant association between 8 q24 rs6983267 polymorphism and susceptibility to prostate cancer. Conclusions The polymorphism of 8 q24 rs6983267 is positively correlated with the susceptibility of prostate cancer.
