摘  要:目的探讨脾多肽对非霍奇金淋巴瘤(NHL)患者浆细胞样树突状细胞(pDC)功能及成熟的影响。方法收集35例NHL患者外周血单个核细胞,采用流式细胞仪分选pDC,用脾多肽刺激pDC 5 d,显微镜观察pDC形态,流式细胞术检测pDC表型,扫描电镜观察pDC超微结构,不同浓度的脾多肽处理24 h后,酶联免疫吸附试验检测pDC上清液中干扰素-α(IFN-α)、肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)水平。以35例同期体检健康志愿者作为对照。结果经不同浓度的脾多肽处理后,pDC的表面突起逐渐明显,随着脾多肽浓度的增加,突起明显变长呈成熟特征;各浓度脾多肽组pDC上清液中IFN-α、TNF-α、IL-6水平均较脾多肽0 mg·L^(-1)组有明显上升,差异有统计学意义(P<0.05),且随着脾多肽浓度的增加而上升明显,各浓度脾多肽组之间差异有统计学意义(P<0.05);各浓度脾多肽组pDC细胞的CD11c、CD80、CD86表达阳性率均较脾多肽0 mg·L^(-1)组有明显上升,差异有统计学意义(P<0.05),且随着脾多肽浓度的增加而上升明显,各浓度脾多肽组之间差异有统计学意义(P<0.05)。结论脾多肽可以有效增强NHL患者pDC的功能,对其分化成熟有很好的促进作用。Objective To investigate the effect of splenic polypeptide on the function and maturation of plasma cell dendritic cells in patients with non-Hodgkin's lymphoma( NHL). Methods Peripheral blood mononuclear cells from 35 patients with NHL were selected. Pulp-like dendritic cells( pDC) were sorted by flow cytometry,and were treated with splenic polypeptide for 5 days. The expression of pDC was detected by immunohistochemistry. The expression of pDC was detected by flow cytometry. The expression of pDC was detected by electron microscope. The expressions of interferon-α( IFN-α),tumor necrosis factor-α( TNF-α) and interleukin-6( IL-6) in the pDC supernatant were detected by enzyme linked immunoadsorption test. The 35 healthy volunteers at the same age were used as controls. Results The expressions of IFN-α,TNF-α and IL-1 in all the pDC supernatant treated with splenic polypeptide groups were significantly higher than those of the no splenic polypeptide group( P〈0.05),and were increased with the increase of spleen peptide concentration. The positive rates of CD11 c,CD80 and CD86 in the pDC supernatant treated with splenic polypeptide groups were significantly higher than those of the no splenic polypeptide group( P〈0.05),and were increased with the increase of spleen peptide concentration. Conclusion Splenic peptide can effectively enhance the function of pDC in the patients with NHL,and it can promote the differentiation and maturation of the patients.
