摘  要:目的探讨成年雄性大鼠经七氟烷麻醉后对海马CA3区Apo E4表达变化的影响。方法 SPF级成年雄性大鼠,行为学测定麻醉前逃避潜伏期后随机分为成实验组Test组(T组,n=30)和对照组Control组(C组,n=30)。实验组调节氧流量1.5L/min+空气流量2.5L/min,吸入3.2%七氟烷麻醉2 h;对照组大鼠吸入氧气1.5L/min+空气2.5L/min共2 h。在麻醉苏醒后2、24、72 h分别再次测定逃避潜伏期。逃避潜伏期测定结束后取脑组织,石蜡切片后行免疫荧光激光共聚焦法检测海马CA3区Apo E4表达量。结果两组大鼠麻醉苏醒后2、24、72 h逃避潜伏期组间比较差异无统计学意义(P>0.05)。T组大鼠苏醒后2、24、72 h海马CA3区Apo E4平均光密度为75.4±25.6、85.3±35.5、85.4±44.0,苏醒后24、72 h与苏醒后2 h比较差异有统计学意义(P<0.01),苏醒后24h与72 h之间比较差异无统计学意义(P>0.05)。C组大鼠苏醒后2、24、72 h海马CA3区Apo E4平均光密度为61.9±12.9、65.3±19.9、65.3±18.9,不同时间点比较差异无统计学意义(P>0.05)。在苏醒后相同时间点T组较C组的海马CA3区Apo E4平均光密度均增高,且差异有统计学意义(P<0.01)。结论成年雄性大鼠经3.2%七氟烷吸入麻醉2 h后,使海马CA3区Apo E4表达持续增高至少72 h,可能是患者术后一定时间内记忆力下降的诱发机制之一。Objective To investigate the effect of the 2 hours sevoflurane anaesthesia on the expression of Apo E4 in the hippocampus CA3 region in the adult male rats. Methods Sixty adult Sprague-Dawley rats were randomly divided into 2 groups after behavioral determination of the escape latency before anesthesia: Test group( group T,n = 30) and Control group( group C,n = 30). According to the three different experimental points( 2,24 and 72 h after the wake of anesthesia),the groups of T and C were respectively divided into three subgroups( T2,T24,T72,C2,C24,and C72,n = 10). 3. 2% Sevoflurane combining with 1. 5 L/min oxygen and 2. 5 L/min O2 were inhaled for 2 hours in group T,while the mixed gas( 1. 5 L/min oxygen + 2. 5 L/min O2) was inhaled for 2 hours in group C. The escape latency was determined at 2 hours,24 hours and 72 hours after the anesthesia was awakened. Take the brain tissue in escape latency after the end of the test were obtained at 2,24 and 72 hours after anesthesia recovery to make immunohistochemical slices. The expression of Apo E4 in CA3 area was detected by immunofluorescence laser confocal method. Results The hippocampus average of optical density of Apo E4 of T2,T24 and T72 respectively was 75. 4 ± 25. 6,85. 3 ± 35. 5,85. 4 ± 44. 0. Compared with T2,average of optical density of Apo E4 was significantly increased in T24 and T72( P〈0. 01). There is no significant difference between T24 and T72( P〉0. 05). Compared with group C,average of optical density of Apo E4 was significantly increased in group T at three different experimental points( P〈0. 01). Conclusion Adult male rats were anesthetized with 3. 2% sevoflurane for 2 hours,the expression of Apo E4 in the hippocampus CA3 region was continuously increased lasting at least 72 hours,which may be one of the inducing mechanisms of memory decline within a certain period of time after the operation.
