摘  要:目的探讨microRNA-144-3p(miR144-3p)对Hep G2细胞侵袭、凋亡的影响及机制。方法选取对数生长期的Hep G2细胞,加入不同浓度的miR144-3p(1μM、10μM、20μM)进行处理,同时设置对照组(加入miRNA无关序列)。培养48 h后检测细胞增殖、侵袭、凋亡情况,并进行肝癌衍生生长因子(HDGF)的表达检测。结果 miR144-3p处理48 h后,miR144-3p能够呈剂量依赖性地抑制细胞增殖,能够呈剂量依赖性地增加细胞凋亡指数,各组比较差异均有统计学意义(P<0.05)。miR144-3p在1~20μM浓度范围能显著抑制Hep G2细胞的穿透能力,呈剂量依赖关系(P<0.05)。miR144-3p能够呈剂量依赖性抑制HDGF的mRNA与蛋白表达水平,其表达水平显著低于对照组(P<0.05)。结论 miR144-3p能抑制肝癌Hep G2细胞的增殖、侵袭与转移,促进细胞凋亡,其作用机制可能与抑制HDGF基因与蛋白表达有关。Objective To investigate the effects and mechanism of effects of MicroRNA-144-3p(miR144-3p) on the proliferation, invasion and metastasis of liver cancer cells. Methods HepG2 cells in logarithmic growth phase were cultured with different concentrations of miR144-3p (1 μM, 10 μM, 20 μM) were selected, while the control group (treated by the nonsense miRNAs) were also selected. The proliferation, invasion and metastasis were detected after cultured 48 h, and the hepatoma derived growth factor (HDGF) were given detection of expression. Results After the cultured of 48 h, miR144-3p showed dose dependent inhibition of cell proliferation, and there were significant difference compared with the control group ( P 〈0.05). And the miR144-3p showed dose-dependent increased in apoptosis index ( P 〈0.05). When the miR144-3p in the concentration range of 1~20 μM , they can significantly inhibit the penetration of HepG2 cells in a dose-dependent manner, and the comparison of each groups were statistically significant ( P 〈0.05). miR144-3p showed the expression level of mRNA and protein in a dose dependent inhibition of HDGF, and the expression level were significantly lower than that of the control group ( P 〈0.05). Conclusion The miR144-3p can inhibit the proliferation, invasion and metastasis of hepatoma HepG2 cells, it can promote cell apoptosis, and its mechanism may be related to the inhibition of HDGF gene and protein expression.
