摘 要:【目的】研究贯叶连翘提取物(Hypericum perforatum L extract)对酒精性肝损伤的保护作用及机制.【方法】用递增法灌胃食用乙醇24周,建立大鼠肝损伤模型,造模同时用贯叶连翘提取物(100、200、300mg/kg)进行灌胃治疗.利用生物显微技术观察肝脏组织结构的变化,全自动生化分析仪检测血清谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)水平,用放射免疫法检测肝脏肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白细胞介素1(interleukin-1β,IL-1β)、白细胞介素10(interleukin-10,IL-10)含量,比色法检测肝脏超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽还原酶(glutathione reductase,GR)活性及丙二醛(malondialdehyde,MDA)含量,用流式细胞术检测细胞凋亡率,用免疫组织化学法及Western-blot检测Bax蛋白的表达情况.【结果】与模型组比较,HPLE治疗后,血清ALT、AST水平降低;肝脏TNF-α、IL-1β、MDA的含量、细胞凋亡及Bax蛋白表达呈降低趋势,IL-10、SOD、CAT、GR水平均显著升高,肝组织损伤明显减轻.【结论】贯叶连翘提取物对酒精性肝损伤有一定的保护作用,机制可能与其可以减少自由基损伤、减轻炎症反应、抑制细胞凋亡有关.【Objective】To explore protection role of the extracts from Hypericum perforatum L.on alcohol hepatic injury.【Method】A hepatic injury model of rat by using edible alcohol was established with increasing dosage for twenty-four weeks.In the process of experimental hepatic injury,the rats were given the extracts at doses of 100,200 or 300 mg/kg by intragastric administration.The changes of liver histological structure were observed by bio-microscopy.The levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum were tested by automatic biochemistry analyzer.And it was detected that the contents of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-10(IL-10)in liver by radioimmunoassay,activities of superoxide dismutase(SOD)and catalase(CAT),glutathione reductase(GR)and malondialdehyde(MDA)content in liver by spectrophotometry,rates of liver apoptosis by flow cytometry,analyzed Bax protein expression in liver by immunohistochemistry and Western-blotting,respectively.【Result】Compared with the model group,the extracts significantly decreased the levels of ALT,AST,TNF-α,IL-1β,MDA,rates of apoptosis and Bax protein expression,while the levels of IL-10,SOD,CAT and GR significantly increased.Further experiments found that the extracts decreased liver injury.【Conclusion】The extracts from H.perforatumhas a protective role against alcohol hepatic injury,due to reduce free radical damage,relieve inflammatory response,and inhibit apoptosis.
