摘  要:目的测定新疆红芪中多糖的含量及其抗氧化活性。方法采用紫外分光光度法,以苯酚-浓硫酸法为显色剂,以5%葡萄糖为对照品绘制标准曲线,测新疆红芪根中多糖含量。以V_C为阳性对照,测定红芪多糖对羟自由基、DPPH自由基的清除能力以及对Cu^(2+)的还原能力。结果新疆红芪多糖浓度在0.019 8~0.058 7mg/mL范围内(Y=15.45 X+0.057 8,r=0.996 9)线性关系良好,新疆红芪中多糖含量为13.77%。对DPPH清除率红芪多糖的IC_(50)为0.208 8μg/mL,V_C的IC_(50)为2.394 0μg/mL。对羟自由基清除率红芪多糖的IC_(50)为0.104 4μg/mL,V_C的IC_(50)为4.788μg/mL。对Cu^(2+)还原能力红芪多糖的IC_(50)为0.069 6μg/mL,V_C的IC_(50)为7.182μg/mL。新疆红芪根中多糖对羟自由基的清除率为81.23%,对DPPH自由基的清除率为83.37%,对铜离子的还原能力为100%。结论本方法快速简便、灵敏度高,适用于新疆红芪中多糖的含量测定。3个抗氧化活性实验结果表明新疆红芪多糖具有较好的抗氧化活性。Objective To determine the content and antioxidant activity of polysaccharides in Hedysarum austrosibiricum b.Fedtsch in Xinjiang.Methods The content of polysaccharide in root of H.austrosibiricum b.Fedtschin Xinjiang root was measured by UV spectrophotometry.Phenol concentrated sulfuric acid was used as chromogenic agent,5% glucose was used to the standard curve.VC was used as a positive control to determine the capacity of H.austrosibiricum b.Fedtsch polysaccharide to scavenge hydroxyl free radical,DPPH radical and reducing Cu^2+ capacity.Results The result showed that the linear relationship was good,when the concentration of H.austrosibiricum b.Fedtsch polysaccharide is in the range of 0.0198-0.0587 mg/mL(Y=15.45 X +0.057 8,r=0.996 9).The content of polysaccharide in H.austrosibiricum b.Fedtsch was 13.77%.The IC50 of polysaccharide in H.austrosibiricum b.Fedtschfor clearance of DPPH was 0.2088μg/mL,and the IC50 of VC was 2.394μg/mL.The IC50 of polysaccharide in H.austrosibiricum b.Fedtschfor scavenging of hydroxyl free radical was 0.1044μg/mL and IC50 of VC was4.788μg/mL.The IC50 of polysaccharide in H.austrosibiricum b.Fedtschfor reducing Cu^2+ was 0.069 6μg/mL and IC50 of VC was 7.182μg/mL.The hydroxyl radical and DPPH radical scavenging rate,and Cu^2+ reduction ability of Polysaccharides in H.austrosibiricum b.Fedtsch was 81.23%,83.37% and100%,respectively.Conclusion The method is rapid,simple and sensitive,and is suitable for the determination of polysaccharides in H.austrosibiricum b.Fedtsch.The results of three antioxidant activities showed that Xinjiang H.austrosibiricum b.Fedtschpolysaccharide had good antioxidant activity.
