定清片对人急性髓系白血病HL-60细胞侵袭力的影响

摘　　要：目的:探讨我院自制中药制剂定清片对急性髓系白血病HL-60细胞侵袭力的影响。方法:制备定清片含药血清,并以不同浓度含药血清对HL-60细胞进行体外干预,Transwell实验检测定清片含药血清对细胞侵袭力的影响,应用免疫印迹法(Western Bloting,简称WB)检测MMP-2、MMP-9基因的蛋白表达量。结果:与空白对照组比较,不同浓度定清片含药血清(5%、10%、20%)作用于HL-60细胞后,不仅对细胞的侵袭能力具有明显抑制作用(P<0.05),还能下调MMP-2、MMP-9基因蛋白表达水平(P<0.05),且随着药物浓度的增加,其抑制力及下调蛋白表达水平作用越强。结论:定清片可抑制HL-60细胞的浸润转移能力,其作用机制可能与抑制细胞侵袭能力及下调MMP-2、MMP-9蛋白表达相关。Objective：To discuss the effects of homemade herbal preparation DingQing tablets on the invasiveness of HL-60 of acute myeloid leukemia（AML）. Methods：Medicated serum of DingQing tablets was prepared, and HL-60 cells were intervened by medicated serum in different concentrations, the effects of medicated serum of DingQing tablets on cellular invasiveness were detected by Transwell experiments, and the protein expressions of MMP-2 and MMP-9 were measured by using western bloting（WB）. Results：Compared with blank control group, after acting on HL-60 cells, medicated serum of DingQing tablets not only showed notable inhibitory effects to cellular invasiveness（P〈0.05）, but also down regulated the protein expressions of MMP-2 and MMP-9（P〈0.05）, as the concentrations of drugs increased, its inhibitory effects and the functions of down-regulating protein expressions were more stronger. Conclusion：DingQing tablets could inhibit invasion and metastasis ability of HL-60 cells, and its mechanism might be related to inhibiting cellular invasiveness and down-regulating the protein expressions of MMP-2 and MMP-9.
